摘 要:目的:探索妊娠期大鼠在雄激素增高环境下对仔鼠出生体重和葡萄糖耐量试验的影响。方法:大鼠孕期不同时间注射雄激素,观察母代妊娠率、流产率、以及对子代OGTT的影响。结果:孕期全程给药组(A组)失去生育力,孕前半程给药组(B组)生育力下降大于孕后半程给药组(C组)(P<0.05)。相应仔鼠组:B'组、C'组D12、D23体重分别与相同天数D'组比较,差异有显著的统计学意义(P<0.01);B'、C'两组间D12和D23体重差异有统计学意义(P<0.05)。B',C'与D'组OGTT比较差异有显著的统计学意义(P<0.01)。结论:孕期高雄激素环境可降低大鼠的生育力。妊娠早期高雄激素造成的生殖毒性作用较强,孕期高雄激素化导致仔鼠体重增加和OGTT异常。Objective: To establish polycystic ovary syndrome ( PCOS ) animal model, and explore the effect of excess androgen exposure during pregnancy period on the body weight and OGTT of SD female rats offspring. Methods : Dehydroepiandrosterone sulfate ( DHEAS ) 6mg/100g was injected( s. e)daily at different time during pregnancy period. The rate of production and miscarriage,the offspring survival rate and weight were determined,and the OGTT were also performed. Results: The mice injected with DHEAS during the whole pregnancy ( group A) lost fertility,and the mice injected with DHEAS in the anterior partial pregnancy period (group B) had more serious fertility injury than group C which was injected in the posterior partial pregnancy period. The offspring body weight of group B or C was heavier than that of the group D(P〈0. 01 ) ,and the offspring body weight of group B was heavier than that of the group C ( P〈0. 05 ). In the OGTT, the offspring glucose levels of group B or C was higher than that of the group D(P〈0. 01 ). There was no statistics differences between the group B and C in OGTT (P〉0. 05 ). Conclusions : Excess androgen exposure during pregnancy period can decrease the rat fertility. Excess androgen stimulation at different degrees and periods will induce heterogeneous of PCOS in the offspring. Excess androgen at anterior partial has stronger effect on reproduction than that of posterior partial during pregnancy. Excess androgen exposure during pregnancy induces increase of body weight and abnormal OGTT in offspring.
