摘 要:目的:建立HPLC法同时测定三七花中鸟苷和腺苷的含量。方法:采用Dikma ODS(200mm×4.6mm,5μm,迪马公司)色谱柱,乙腈-水二元梯度洗脱,流速1.0mL·min-1,检测波长254nm,柱温30℃。结果:鸟苷和腺苷分别在17.2~103.2μg·mL-1(r=0.9995,n=6)和15.6~93.6μg·mL-1(r=0.9997,n=6)范围内呈良好线性关系;平均回收率分别为97.6%~99.7%(n=3)和98.1~99.2%(n=3)。结论:本测定方法简便、快速、准确,为三七花质量评价提供了可靠方法。Objective:An RP-HPLC method was developed to determine the contents of guanosine and adenosine simultaneously in flower buds of Panax notoginseng.Methods:The analysis was performed on a C18 column with gradient elution of acetonitrile and water at a flow rate 1.0 mL·min -1.The UV detection wavelength was 254 nm,and the column temperature was set at 30 ℃.Results:The linear range was 17.2-103.2 μg·mL-1 (r=0.9995,n=6) for guanosine and 15.6-93.6 μg· mL -1 (r=0.9997,n=6) for adenosine.The average recoveries of guanosine and adenosine were 97.6%-99.7% (n=3) and 98.1%-99.2% (n=3),respectively.Conclusion:The method was found to be simple and accurate for simultaneous analysis of guanosine and adenosine in flower buds of Panax notoginseng,and provided a reliable way for evaluating the quality of it.
