摘 要:本研究旨在初步探讨IFN-γ不同基因型(AA,AT和TT)髓系祖细胞在1,4-苯醌(1,4-BQ)作用下造血损伤的影响是否存在差异。采用PCR对36例足月分娩脐带血(cord blood,CB)标本干扰素-γ(interferon-γ,IFN-γ)基因+874位点单核苷酸多态性(SNP)进行检测筛选,依据筛选结果将标本分为3组(AA组,AT组,TT组),使用完全甲基纤维素培养基(METHOCULTTM GF H4434)培养单个核细胞(mononuclear cells,MNC),用不同浓度的1,4-苯醌(1,4-Benzoquinone,1,4-BQ)处理,进行集落形成单位(colony forming unit,CFU)检测。结果表明,收集的36份CB中IFN-γ+874AA,AT和TT基因型分布频率分别为5.56(,88.89(,5.56(。在不同浓度1,4-BQ(0,5,10,20μmol/L)作用下IFN-γ+874A/T不同基因型(AA、AT、TT)组内比较集落生长能力,结果显示差异有统计学意义,p值分别为:pAA=0.033,pAT=0.009,pTT=0.001,均小于0.05。1,4-BQ浓度?5μmol/L时,表现出细胞毒性,而且随着1,4-BQ浓度的增大,集落计数就越少,且形态也越小。同一浓度1,4-BQ作用下,IFN-γ+874A/T不同基因型(AA、AT、TT)各组集落生长能力未表现出明显差异。AT组内标本3和TT组内标本2较其它标本集落计数明显少。结论:1,4-BQ对IFN-γ+874AA、AT、TT不同基因型髓系祖细胞生长均有毒性作用且呈浓度依赖性,但在同一浓度1,4-BQ作用下,IFN-γ+874A/T不同基因型(AA、AT、TT)髓系祖细胞集落生长情况未表现出明显的不同。This study was aimed to investigate the effect of 1,4-benzoquinone ( 1,4-BQ ) on growth of myeloid progenitor cells with IFN- γ different genotypes and to compare its differences. Polymerase chain reaction (PCR) was used to amplify the polymorphism gene segment of IFN-γ + 874 A/T in 36 cord blood(CB) specimens. The specimens were divi-ded into three groups (AA, AT and TT group). MNCs were planted on complete methylcellulose medium containing different concentrations of 1,4-BQ. The colony-forming units (CFU) were assayed, the differences of colony growth in specimens with different genotypes ( AA, AT and TT) under 1,4-BQ exposure were analyzed. The results showed that frequencies of AA, AT and TT genotypes were 5.56%, 88.89% and 5.56% in the 36 CB samples respectively. Comparing colony numbers of IP-N-γ + 874 AA, AT and TT genotype indicated that there was significant difference (PAA = 0. 033, PAT = 0. 009, pTT = 0. 001, 〈 0. 05 ). Significant cytotoxicity was observed after exposure to concentrations of 1,4-BQ ≥ 5 μmol/L. Cytotoxic response of 1,4-BQ was dose-dependent. Under the same concentration of 1,4-BQ, there were no significant differences in capacity of cell colony growth between 3 groups (AA, AT and TT) . Colony numbers of specimen No 3 in AT group and specimen No 2 in TT group were less than those of other specimens significantly. It is concluded that the hematopoietic myeloid progenitor cells cultured in the presence of 1,4-BQ show a dose-dependent cytotoxic response, but there are no significant differences in colony growth of IFN-γ different genotypes (AA, AT and TT) under the same concentration of 1,4-BQ.
