摘  要:高脂饲料喂养20周的sD肥胖大鼠呈胰岛素抵抗特征,免疫荧光双标记显示内源性大麻素受体1(CB1R)主要表达于胰岛D细胞,免疫组化和实时定量PCR结果显示,高脂喂养组胰腺CB1R蛋白(6002.8±855.5vs3712.8±769.6,P〈0.01)及mRNA(对照组的16.7倍,P〈0.05)表达均高于对照组,提示肥胖时胰岛β细胞CB1R表达量上调可能导致胰岛素抵抗。Obese rats manifested the characteristics of insulin resistance after high-fat-diet feeding for 20 weeks. Double immunofluorescence showed that endocannabinoid receptor 1 (CB1R)was greatly expressed in pancreatic β-cells. The results of immunohistochemistry and real-time quantitative PCR revealed that CB1R protein (6 002.8±855.5 vs 3 712.8±769.6 ,P〈0.01 ) and mRNA( 16.7 folds of control group,P〈0.05 ) expressions in high-fat group were higher than those of control group, suggesting a role of CB1R in high-fat-diet induced insulin resistance.
