摘  要:金属硫蛋白是一种普遍存在于生物体内的低分子量、半胱氨酸含量丰富、易于被外界刺激诱导的金属结合蛋白。采用表达序列标签(EST)法,首次获得了文蛤金属硫蛋白(Mm-MT)的全长cDNA序列,序列全长657bp,开放阅读框长度为231bp,可以编码76个氨基酸。编码的氨基酸中半胱氨酸含量丰富,达27.6%,含有软体动物等无脊椎动物金属硫蛋白的特征序列。序列特征分析表明,该序列具备金属硫蛋白的典型特征,是金属硫蛋白家族成员。Metallothioneins (MT) are low molecular weight, cysteine-rich, inducible metal-binding proteins found in a variety of organisms. MTs have many important biological functions acted as a protective role against excess reactive heavy metal ions, free radical scavengers and reservoir of essential metals that can be donated to other metalloproteins. The full-length eDNA sequence of metal- lothionein (named as Mm-MT) gene in the hard clam (Meretrix meretrix) was identified from M. meretrix eDNA library using ex- pressed sequence tag (EST) method. The full-length eDNA of Mm-MT is of 657 bp and an open reading frame (ORF) of 231 bp en- coding a polypeptide of 76 amino acids. The contents of cys in the -Mm-MT amino acid sequence is 27.6%. Mm-MT is characterized by the conserved features of invertebrate's MTs. The result of sequence analysis indicates that Mm-MT is the member of metallothionein family. This research will provide basic information for evolution and function study of marine shellshef metallothionein.
