摘 要:[目的]通过比较不同药物干预对多药耐药基因1(MDR1)的影响,观察难治性癫痫大鼠脑内MDR1mRNA的表达和分布,探讨难治性癫痫多药耐药可能的分子病理机制,及中药复方柴贝止痫汤对其表达的影响。[方法]以不同药物干预下,观察癫痫发作的平均持续时间、癫痫发作级别,以判定、分析药物的疗效。采用荧光实时定量聚合敏链反应(PCR)的方法,分析、比较多药耐药基因MDR1在各组大鼠海马和皮质部位的表达和分布。[结果]与正常组大鼠比较,各致痫组MDR1mRNA表达含量均有明显增高,具有统计学意义(P<0.05)。[结论]癫痫的反复发作可以诱导慢性难治性癫痫大鼠海马和皮质MDR1mRNA的表达,中药复方柴贝止痫汤具有一定抑制MDR1药物转运体功能。[Objective] To investigate the effects of Chaibei Zhixian decoction, a Chinese herbal compound, on the expression and distribution of multiple drug resistance gene 1 (MDR1) mRNA in intractable epileptic rats, to explore the possible molecular pathogenesis mechanism of the intractable epilepsy. [Methods] The average duration of seizures and the Racine's stage of epileptic seizure under different drug's intervention were observed to judge and analyze their effects. Real-time PCR was used to compare the expression and distribution of MDR1 in the hippocampus and cerebral cortex of the rats. [Results] Compared with the normal group, the expression of MDR1 mRNA was significantly elevated in all epileptic groups. ( P〈0.05). [Conclusion] Spontaneous recurrent seizure can induce the expression of MDR1 in the hippocampus and cerebral cortex in chronic intractable epileptic rats. The Chinese herbal compound Chaibei Zhixian Tang had some efficacy in inhibiting MDR1 drug transporters.
