膝骨性关节炎患者滑膜和滑液中基质金属蛋白酶-3表达水平的研究

摘　　要：目的比较骨性关节炎和半月板损伤患者关节滑膜和滑液中基质金属蛋白酶-3的表达,探讨OA的发病机制。方法收集骨性关节炎患者关节滑膜和滑液43例,以单纯半月板损伤患者的关节滑膜和滑液45例为对照;采用免疫组织化学染色法,观察骨性关节和半月板损伤患者关节滑膜MMP-3阳性表达强度;采用ELISA法检测两种滑液标本基质金属蛋白酶-3的水平。结果MMP-3在OA滑膜组织中的表达与对照组相比差异有统计学意义(P(0.01);MMP-3在OA滑液中的水平与对照组相比差异也有统计学意义(P(0.05);MMP-3在OAX线分级为I度的关节滑膜和滑液中的表达和II度相比无统计学意义(P>0.05)。结论MMP-3在OA发生发展中扮演着重要的角色。Objective To investigate the level of MMP-3 in synovium and synovial fluid of patitents with primary osteoarthritis （OA）,and study the role of MMP-3 in OA.Methods The synovium and synovial fluid of knee joint were collected from 43 subjects with meniscus injury,45 adult patients with OA.The expression of MMP-3 in synovium was analyzed by immunohistochemistry staining.The level of MMP-3 was determined by ELISA.Results The intensity of positive cells with expression of MMP-3 in synovium was significantiy higher in patients OA than in subjects with meniscus injury （P0.01）.The level of MMP-3 in patients with OA was significantiy higher than in subjects with meniscus injury in synovial fluid （P0.05）.The level of MMP-3 in synovium and synovial fluid was not statistical significance in patients between one degree and two degree typed with X radioactive ray.Conclusion MMP-3 plays significant role in the pathogenesis of OA.
