摘  要:目的将过期的血小板制成微粒型血小板制剂,研究其与创面愈合及修复的作用关系。方法将过期的单采血小板冰冻干燥制成微粒型血小板制剂,制备的干粉质量为约0.28g/ml;分别采用ELISA的方法检测常规(液态)血小板与微粒型血小板制剂(复溶成液态)中VEGF、TGF-β1、PDGF的含量;将微粒型血小板制剂应用于全层皮肤缺损的小鼠背部皮肤创面,观察该制剂是否具有促进伤口愈合的功能;把小鼠随机分为生理盐水组、血浆组和微粒型血小板制剂组(每组每时相点6只),每只小鼠右侧均为生理盐水对照组,两组给药50μl(干粉用生理盐水复溶成液态)1次/d,观察伤后各组各时相点创面愈合情况,并应用免疫组化的方法检测各组术后5、7、10、14d创面皮肤组织CD31的表达情况。结果常规液态血小板和微粒型血小板制剂(液态)组中VEGF的含量,无统计学差异(t=-1.616,P>0.05),而微粒型血小板制剂组中TGF-β1、PDGF的含量较相应的常规液态血小板为高(t=-2.269、2.338,P均<0.05)。微粒型血小板制剂组在小鼠创伤后d5、7、10创面面积较盐水对照组和血浆组有明显的减小(t分别=2.997、3.427、10.78,t=2.599、2.331、6.407,两组P均<0.05);d5、7、10创面收缩率与盐水对照组相比有明显的提高(t=2.542、3.161、5.884,P<0.05),d7、10与血浆组相比,t=2.177、4.351,P均<0.05;实验组在创伤后d5、7、10创面组织CD31的积分光密度与盐水对照组相比有明显升高(t=-2.339、-3.553、-4.941,P<0.05)。d7、10与血浆组比较,t=-2.373、-2.412,P均<0.05。结论微粒型血小板干粉制剂中TGF-β1、PDGF较常规血小板中增高;其对小鼠皮肤创面的修复和愈合有促进作用。Objective To study the effect of microparticle platelet frozen-powder prepared from outdated platelets on wound healing in animal models. Methods Microparticle platelet frozen-powder was prepared by repeated freeze-thawing and lyophillzing outdated platelets. The specific gravity of this powder was 0. 28 g/ml. Levels of VEGF, TGFβ1 and PDGF in apheresis platelets and mieroparticle platelet frozen-powder were measured and evaluated by ELISA. The mouse model with skin wounds on the back was used to study the effect of the powder on wound healing. Kunming mice were randomly, divided into control group, plasma group, and mieroparticle platelet frozen-powder group, with six mice in each group. A 1.2 centimeter-diameter full-thickness wound was made on each side of the mouse mid-back. The treatment was applied to each wound once daily for 14 eousecutive days after wounding. The wound area, the percentage of wound contraction and CD31 expression on days 5, 7, 10, and 14 after wounding were also measured. Results Levels of VEGF were not significantly different in fresh apheresis platelet and mieroparticle platelet frozen powder ( P 〉 0. 05 ). Levels of TGF-β1 and PDGF in mleropartiele platelet frozen powder were higher than those in fresh platelet ( P 〈 0.05 ). Wound area was decreased in the micropartiele platelet frozen powder group, compared with those in the saline control group and plasma control group on days 5, 7 and 10 after wounding( P 〈 0 05 ). The percentage of wound contraction was enhanced in the micropea'tide platelet frozen powder group compared with those in the saline control group and plasma control group on days 5,7 and 10 after wounding( P 〈 0. 01, P 〈 0. 05 ). CD31 expression was higher in mieropartiele platelet frozen powder group compared with those in saline control group and plasma control group on days 5, 7 and 10 after wounding( P 〈 0. 01, P 〈 , 0. 05 ) .Condusion Levels of TGF-β1 and PDGF in micropartiele platelet frozen powder were higher, which can impr
