摘 要:目的通过观察丰白散对慢性阻塞性肺疾病(COPD)模型大鼠γ-谷氨酰半胱氨酸合成酶(γ-GCS)与超氧化物歧化酶(SOD)的影响,探讨丰白散治疗COPD的作用机制。方法采用烟熏加气管内滴加脂多糖方法复制COPD模型,用苏木素-伊红(HE)染色评估各组大鼠肺组织病理改变,逆转录聚合酶链式反应(RT-PCR)检测γ-GCSmRNA表达水平,用黄嘌呤氧化酶法测定SOD活力。结果模型组、丰白散组、沐舒坦组大鼠肺组织出现了符合人COPD的病理改变,COPD模型复制成功;与正常组相比,模型组大鼠肺组织γ-GCSmRNA表达水平、SOD活力均明显降低,两者比较差异有统计学意义(P<0.05);与模型组相比,丰白散组肺组织γ-GCSmRNA表达水平、SOD活力均增强,差异有统计学意义(P<0.05)。结论丰白散能增强COPD模型大鼠γ-GCS表达及SOD活力,纠正氧化/抗氧化失衡,增强抗氧化能力,这可能是丰白散治疗慢性阻塞性肺疾病的作用机制之一。Objective To observe effects of Fengbaisan decoction on the expression of gamma-glutamylcysteine synthetase(γ-GCS) and the activity of superoxide dismutase (SOD) in rats with chronic obstructive pulmonary disease, and study the therapeutic effects and the potential mechanism of the decoction. Methods COPD model was established by modified method of combining fumigation and lipopolysaccharide (LPS) intra-tracheal dripping; Lung tissue sections stained by HE were observed to study the morphological changes; Reverse transcription polymerase chain reaction (RT -PCR) was used to evaluate the mRNA level of gamma-glutamylcysteine synthetase (γ-GCS); superoxide dismutase (SOD) was detected by adopting purine oxidase method. Results The typical pathologic change of emphysema was emerged in three groups of COPD model of rats; Compared with the normal control group, the expression of γ-GCSmRNA and the activity of SOD in COPD model group were decreased significantly, there were significant difference(P〈0.05); the expression of γ-GCSmRNA and the activity of SOD in the fenbaisan Treatment group was increased markedly as compared to the COPD model group, there were statistical difference(P〈0.05). Conclusion Fenbaisan decoction can strengthen the expression of γ-GCS and the activities of SOD, and correct the oxidant/antioxidant imbalance, which may be one of the mechanism of the decoction to treat the chronic obstructive oulmonarv disease.
