摘 要:目的观察银杏叶提取物(EGB)对Aβ25-35所致阿尔茨海默病(AD)模型大鼠的学习记忆能力及脑内海马神经元c-Rel和Bcl-xl表达的影响。方法采用双侧海马注射Aβ25-35制备AD大鼠模型,采用Morris水迷宫检测EGB对模型大鼠认知功能的的影响;通过病理形态学观察EGB对模型大鼠的脑保护作用;采用免疫组化法观察EGB对模型大鼠海马神经元c-Rel和Bcl-xl的表达。结果AD组与假手术组(SC)比较,每天隐匿平台逃避潜伏期均明显增长(P<0.05),海马CA1区Bcl-xl和c-Rel阳性细胞光密度明显减少(P<0.05);EGB(150和300mg/kg)组与AD组比较,空间学习记忆能力明显改善(P<0.05),海马CA1区Bcl-xl和c-Rel阳性细胞光密度明显增高(P<0.05)。但EGB1(150mg/kg)组与EGB2(300mg/kg)组比较,每天隐匿平台逃避潜伏期和海马CA1区Bcl-xl和c-Rel阳性细胞光密度值差异无统计学意义(P>0.05)。结论EGB能通过增强c-Rel及Bcl-xl的表达,抑制海马神经元的凋亡,改善AD大鼠学习记忆能力。Objective To study the effects of the extracts of ginkgo biloba leaves on the expression apoptosis related gene c-Rel and Bcl-xl of the hippocampus neurons in rat model of Alzheimer's disease induced by β-amyloid (Aβ25-35 )and it's learning and memory ability. Methods The AD model of rats was established by injecting Aβ25-35 into the CAl district of both hippocampus. The effect of the extracts of ginkgo biloba leaves on learning and memory retention disorder in the model rats was studied by the test of Morris water maze; the brain protection of the extracts of ginkgo biloba leaves in model rats was observed through path morphology changes ; the expression of c-Rel and Bl-xl of hippocampus neurons was detected by immunohistochemistry method. Results The escape latency of the place navigation of the AD group was significantly longer than the sham injection group(P〈0. 05) ,Aβ25-35 could reduce the optical density of positive ceils of c-Rel and Bcl-xl(P〈0. 05)of hippoeampus area;The AD rats received EGB( 150and 300mg/kg)showed significant shorter escape latency than AD rats (P〈0.05), EGB could increase the optical density of positive cells of c-Rel and Bcl-xl( P〈0. 05 );but comparing EGB (150mg/kg)with EGB (300mg/ kg) ,the escape latency of the place navigation and the optical density of positive ceils of c-Rel and Bcl-xl was not significantly different (P〉0. 05 ). Conclusion The extracts of ginkgo biloba leaves (150and 300mg/kg)enhanced the expression of c-Rel and Bcl-xl to restrain the apoptosis of the hippocampus neurons, improve the learning and memory of AD rats.
