摘 要:目的探讨四氯化碳(CCl4)一过性处理对巴马香猪肠道菌群的影响,评估CCl4诱导处理的肝损伤动物模型能否用于肠道微生态研究。方法选取3头雄性巴马香猪,一次性按每千克体重腹腔注射0.25 ml[0.25ml/(kg.bw)]40%CCl4橄榄油溶液。于注射12 h之后的第1、2、3、4、7和10天连续采集粪便,以注射CCl4溶液之前所采集粪便样作对照。分别提取粪样总DNA,采用变性梯度凝胶电泳技术(DGGE)对CCl4一过性处理后巴马香猪肠道菌群的多样性变化进行动态监测。结果PCR-DGGE法分析表明,巴马香猪肠道菌群组成在注射CCl4前后差异无显著性(P>0.05)。相似性聚类分析结果表明,菌群组成在注射CCl4前后相似度在65%以上。结论CCl4一过性处理后巴马香猪肠道菌群组成未发生显著变化。表明CCl4处理诱导的肝损伤动物模型可以用来研究相关疾病与肠道菌群之间的关系。Objective To investigate the effects of Carbon tetrachloride transient treatment on intestinal microflora of Bama minipigs,and evaluate whether the liver-damaged animal model introduced by CCl4 could be applicable for the study of intestinal flora.Method Three male Bama minipigs were intraperitoneally injected with 40% CCl4 olive oil solution.Injection volume was 0.25 ml/(kg·bw).Fecal samples were collected on the 1^th,2^nd,3^rd,4^th,7^th and 10^th days after the injection of CCl4 solution.The fecal samples of control group were collected before the injection.The genomic DNA of bacteria was extracted from each sample.The variety of intestinal microbiota in fecal samples was tested by PCR-DGGE.Result The PCR-DGGE results showed no significant difference in the intestinal microflora composition before and after injection of CCl4(P〉0.05).Similarity cluster analysis results showed that the similarity of flora composition before and after injection of CCl4 was more than 65%.Conclusion There was no significant difference in the composition of intestinal microflora of Bama minipigs transiently treated with CCl4.The liver-damaged animal models introduced by CCl4 can be used to study the relationship between intestinal flora and related diseases.
