免疫组织化学方法诊断猪圆环病毒病

摘　　要：以猪圆环病毒Ⅱ型ORF2(PCV2-ORF2)重组蛋白为抗原,按常规免疫方法免疫家兔,制备兔抗猪圆环病毒Ⅱ型ORF2重组蛋白的IgG,以此IgG为第一抗体,采用免疫组织化学二步法,对临床疑似猪圆环病毒病例及RT-PCR确诊为阳性猪圆环病毒病例的肺脏组织切片进行检测。试验结果证明,该免疫组织化学方法具有良好的特异性、直观性,简单易行,可以对猪肺巨噬细胞中的存在的PCV2病毒抗原进行准确的定性和定位诊断。To prepare the immunoglobulin G （IgG） of rabbit anti-Porcine Circovirus-Ⅱ ORF2 recombination protein, we immunized rabbit through conventional methods and used porcine circovirus type Ⅱ （PCV2-ORF2） as the antigen. Detection was carried on the tissue slices of lung resource from clinical suspected cases of PCV and the positive cases diagnosed by the RT PCR, the IgG was taken as the first antibody, by adopting two-step immunohistochmical method. The result indicated that the immunohistochemistry assay has good characteristics of specificity, intuitive and convenient, and could exactly diagnose for qualitation and localization of virus antigen of PCV2 in macrophage from pig lung.
