丽江云杉胚性愈伤组织石蜡制片中的整块染色与切片染色方法比较

摘　　要：以丽江云杉胚性愈伤组织为材料,运用整块染色方法与切片染色方法对其石蜡制片的效果进行比较。结果表明,两种染色方法制成的切片在观察效果上无明显差异,均能对丽江云杉的胚性愈伤组织的细胞核、淀粉粒染色,且各部分的着色均匀一致,染色效果佳。两相比较,整块染色法的操作步骤略为简化,能在一定程度上减轻切片污染、皱褶和蜡片脱落现象的产生。The embryonic callus of Picea likiangensis was used as the material for two methods of staining, entire staining and section staining with which olefin section was made. It showed that there was no significant difference in the two methods in the staining of nucleus and starch particles. The quality of the staining was good and homogeneous. It suggested that the entire staining be simply manipulated and reduce pollution and wrinkles and dropping of the sections.
