QTL Analysis of Sheath Blight Resistance in Deepwater-rice Chihe(Oryza sativa L.)

摘  要:以高抗纹枯病的深水稻品种赤禾及感病品种Lemont为材料,构建了F2群体,以牙签嵌入法对该群体128株单株进行纹枯病抗性鉴定。采用105个均匀分布于水稻12条染色体上的SSR多态性标记,构建该群体的分子标记连锁图,进行数量性状位点(QTL)分析。以LOD值2.5为阈值,在全基因组范围内搜索,初步检测到3个主效抗病QTLs,暂命名为qSB-2、qSB-5和qSB-11,分别位于第2、5和11染色体上,各自能解释表型抗性变异的17.17%、14.7%和27.78%,其中qSB-5来自抗病亲本赤禾,qSB-2,、qSB-11来自感病亲本Lemont。In this study, Chihe, a deep-water rice variety resistance to sheath blight, and an indica variety Lemont and 128 plants of their crossing F2 population were investigated for Sheath blight resistance using the toothpick method. A molecular linkage map of the F2 population was constructed with 105 pairs SSR markers which distribute equally on the rice 12 chromosomes. Quantitative trait loci (QTL) analysis was undertaken based on the molecular linkage map using the QTL Cartographer version 2.0. Three sheath blight resistant QTLs( qSB-2, qSB-5, and qSB-11 ) were identified by genome-wide analysis method with LOD score of 2.5, and located on chro~ 2, 5 and 11, respectively. Resistant alleles of qSBR-2 and qSBR-5 were derived from the resistant parent Chihe, whereas resistant allele of qSBR-11 from susceptible parent Lemont. The qSBR-2, qSBR-5 and qSB-11 explained 17.17 %, 14.7 % and 27.78 % of the phenotypic variance, respectively.
