摘 要:目的研究吲哚-3-甲醇(I3C)的一种新衍生物I3C-6对人类宫颈癌HeLa细胞生长的影响。方法采用MTT法和体外划痕法检测HeLa细胞的生长和浸润;采用流式细胞术检测对细胞周期的影响;采用DNA梯带凝胶电泳法检测细胞凋亡,采用Westernblot方法检测蛋白表达。结果I3C-6呈现剂量依赖性抑制HeLa细胞的生长,IC50剂量约为10μmol/L。I3C-6处理HeLa细胞后出现显著的细胞凋亡峰(SubG1峰)。DNA凝胶电泳结果也提示I3C-6处理的细胞出现明显的凋亡DNA片段。I3C-6可下调Bcl-2蛋白的表达,并上调Bax蛋白的表达。结论I3C-6可以抑制人类宫颈癌HeLa细胞生长,这与其诱导的细胞凋亡、细胞周期抑制以及凋亡蛋白调节有关。I3C-6可能是一种有效的抗宫颈癌药物和辅助治疗药物。Objective To study the impact of indole-3-carbinol-6 (I3C-6) on the growth,the migration,the involved mechanisms and the radiosensitivity of the cervical cancer HeLa cells. Methods HeLa cells were harvested for cell survival evaluation by MTT assay; migration by scratch adhesion test; cell cycle analysis by flow cytometry assay; apoptosis by DNA gelelectrophoresis assay; protein expression by Western blot; radiosensitivity by clone formation method. Results I3C-6 inhibited the growth and the migration of HeLa cell in a dose-dependent manner.It blocked cell cycle progression at apparent G1 arrest. Apoptosis in duction was also observed as 180~200 bp DNA ladder appearing on agarose gel. I3C-6 up-ragulated Bax,and down-regulated Bcl-2.Radiosensitivity were significantly increased by I3C-6. Conclusion I3C-6 can significantly inhibit HeLa cell growth and migration. Apoptosis induction may be the mechanism for I3C-6 inhibition of cell growth. Radiosensitivity is also significantly increased by I3C-6. These results indicates that the preclinical rationale for clinical protocols of honokiol improves the cervical cancer chemotherapy.
