摘 要:目的以鼠麴草的叶为材料,研究鼠麴草DNA的提取方法及对其进行随机扩增多态性DNA(RAPD)的分析。方法采用略改进的十六烷基三甲基溴化铵(CTAB)法、高盐低pH法、木本法和十二烷基硫酸钠(SDS)法分别提取鼠麴草叶的基因组DNA。通过RAPD分析所提取的DNA,比较所用的提取方法。结果CTAB法得到DNA的A260/A280为1.937,浓度为992.25μg/ml,优于其他3种方法。结论CTAB法是4种方法中最佳的方法。Objective To study the methods for extraction of genomic DNA from leaves of Gnaphalium affine D.Don and to analyse its random amplified polymorphism DNA (RAPD) Methods Little improved cetyltrimethyle ammonium bromide (CTAB) method,woody method,low pH medium with high salt and sodium dodecyl sulfate (SDS) method were used to extract genomic DNA from leaves of Gnaphalium affine D. Don. The isolated DNA samples were able to could be used directly by RAPD analysis and then we were able to compare the four methods. Results The CTAB method was the best with comparing with yield and quality of four extraction methods,Its A260/A280 ratio and concentration were 1.937 and 992.25 μg/ml respectively. Conclusion CTAB methods is considered to be a optimal technique among the four methods.
