摘 要:目的:研究体外培养的小鼠树突状细胞(dentritic cells,DCs)和抗原负载树突状细胞在肿瘤模型鼠体内的分布和形态,为肿瘤的生物学治疗提供形态学基础。方法:分离和培养DC,制备B16黑色素瘤细胞抗原,进行共培养,即为抗原负载的DC,Brdu标记DC和抗原负载的DC,建立B16黑色素瘤小鼠模型,于瘤周围皮下注射Brdu标记的DC和抗原负载的DC。应用光镜、免疫组化方法和透射电镜观察DC和抗原负载DC在肿瘤模型鼠体内的分布和形态。结果:免疫组化染色显示Brdu标记的抗原负载DC与DC比较,体积较大。实验组Brdu标记的DC和抗原负载DC分布的数密度和面密度,分别与对照组比较,有显著差异(P<0.01)。电镜下抗原负载DC细胞与DC比较体积较大,核有切迹,细胞表面的突起较粗大弯曲,形态较成熟。结论:抗原负载DC比DC更易集聚于肿瘤组织周围,推测抗原负载DC比DC可能诱导抗肿瘤效应更强。Objective:To investigate the distribution and morphology of the cultured dentritic cell(DC) and antigen-loaded DC in tumor mouse model in Vivo,provide morphological basis for the tumor biological therapy. Methods:DCs were separated and cultured. antigen was extracted from B16 melanoma cells by method of freeze thawing. They were co-cultured. The co-cultured cells were antigen-loaded DC. DC and antigen-loaded DC were marked by Brdu. The mouse model of B16 melanoma was established. Brdu-marked DC and antigen-loaded DC were injected into the tumor-surrounding tissues. The distribution and morphology of DC and antigen-loaded DC in tumor mouse model in Vivo was observed by optical microscopy,electron microscopy and immunohisochemistry. Results:The antigen-loaded DC cells were bigger than the DC cells. The number(area) density of Brdu-marked DC and antigen-loaded DC was significantly higher in the model group than that in the controls respectively(P〈0.01) . Under electron microscopy,the antigen-loaded DC cells were bigger than the DC cells and notch which always appears on cellular nucleus was observed. The protrusions on the surface of antigen-loaded DC cells were more grossus and curved than that of DC,and their morphology was more mature. Conclusion:The antigen-loaded DC cells were more accumulated to the tumor-surrounding tissues compare with DC. These suggested that the role of against tumor induced antigen-loaded DC was stronger than that of DC.
