摘  要:目的:探讨壳聚糖/I型胶原复合物作为软骨组织工程支架材料的物理性能和生物学特性。方法:将壳聚糖溶液与I型胶原溶液混合,梯度冷冻后通过冷冻干燥法制备成复合支架。以纯壳聚糖支架材料作为对照。扫描电镜检测支架材料的形貌,检测支架材料的孔径、孔隙率和吸水率。酶消化法培养滑膜细胞,将第3代滑膜细胞接种于支架材料中,检测滑膜细胞在支架材料中的粘附情况,并通过噻唑蓝(MTT)法检测支架材料对滑膜细胞增殖的影响。结果:制备的支架材料呈孔隙分布均匀的海绵状。支架材料的平均孔径为(119.32±38.67)μm,孔隙率为(91.40±1.41)%,吸水率为(69.10±5.61)水g/支架干重g。滑膜细胞在壳聚糖/Ⅰ型胶原支架材料中的粘附率为98.73%,并且该支架材料对滑膜细胞的增殖具有明显促进作用。结论:壳聚糖/Ⅰ型胶原支架材料可作为细胞载体应用于软骨组织工程。Objective: To investigate the characteristics of chitosan/collagen type Ⅰ (CS/COL-Ⅰ )copolymer as a scaffold for cartilage tissue engineering. Methods: CS/COL-Ⅰ copolymers were synthesized through the method of freeze-drying. The morphological characteristics were examined by means of scanning electron microscope; The pore size and porosity as well as water-binding capacity were evaluated. Passage 3 synovial cells were cultured onto the CS/COL- Ⅰ scaffolds to explore the cell attachment capacity and the proliferation of synovial cells on the scaffolds. Results: The synthesized scaffolds exhibited interconnected porous copolymer with pore size of (119.32±38.67) μm, porosity of (91.4±1.41)%, and water-binding capacity of (69.1±5.61) g (water)/g (dry scaffold). After passage 3 synovial cells being cul- tured onto CS/COL-Ⅰ scaffolds, the cell attachment capacity was evaluated as 98.73%, and the proliferation of synovial cells on the scaffolds was increased. Conclusion: It is suggested that the synthesized CS/ COL- Ⅰ copolymers could be served as a candidate scaffold for cartilage tissue engineering.
