摘  要:目的探讨盐酸戊乙奎醚(penequinine hydrochloride,PHC)对百草枯(paraquat,PQ)所致大鼠急性肺损伤(acute lung injury,ALI)的保护作用及其可能的机制。方法雄性SD大鼠54只随机分为对照组(n=6)、PQ组(n=24)和PHC组(n=24),后两组均再分6、24、48、72h四个亚组(n=6)。PQ组和PHC组大鼠腹腔注射PQ(25mg/kg),对照组腹腔注射等容的生理盐水;PHC组在PQ注射后2h再腹腔注射PHC(0.3mg/kg),PQ组腹腔注射等容的生理盐水。用ELISA法分别测定各时间点各组大鼠血浆和支气管肺泡灌洗液(bronchoalveolar,BALF)中丙二醛(malondialdehyde,MDA)和超氧化物歧化酶(superoxide dismutade,SOD)的含量;测定肺组织湿,干重比(W/D);光镜下观察肺组织结构变化;并采用免疫组化方法测定大鼠肺组织核因子-κB(NF-κB)p65的表达。结果与对照组比较,PQ组各时间点血浆及BALF中MDA水平明显升高(P〈0.01)、血浆中SOD水平明显下降(P〈0.05);BALF中SOD水平6、24h时下降(P〈0.05),但48h后升高(P〈0.01);PHC组与PQ组比较,血浆和BALF中MDA含量明显下降(P〈0.05),SOD含量明显升高(P〈0.05);光镜下观察:PHC组各组肺组织病理变化较PQ组减轻;PHC组肺组织W/D值及NF-κB p65的表达较PQ组明显下降(P〈0.05)。结论PHC对PO中毒所致的大鼠ALI具有保护作用。Objective To study the protective effects of penequinine hydrochloride on paraquat-induced acute lung injury in rats. Methods Fifty-four male SD rats were all injected intraperitoneally with paraquat (25 mg/kg) to induce lung injury (PQ groups), or with the same volume of normal saline (control group). Penequinine hydrochloride (0.3 mg/kg) was injected intraperitoneally 2 h later in half amount rats of the study group, and the same volume of normal saline was injected in another half amunfs (PHC group). Rats were sacrificed 6, 24, 48 or 72 h after penequinine hydrochloride or saline injection. The malondialdehyde (MDA) level and superoxide dismutade (SOD) activity in both plasma and bronchoalveolar lavage fluid (BALF) were detected, respectively. Expression of nuclear factor kappa B (NF-κB) p65 in lung was measured with immunohistochemistry staining, and lung wet/dry weight ratio (W/D) was calculated. Results MDA level in PQ group increased significantly both in plasma and BALF (P〈0.01), SOD activity decreased both in plasma and BALF (P〈0.05) but increased significantly in BALF 48h later (P〈0.01). After injection of penequinine hydrochloride, MDA decreased and SOD activity increased significantly in both plasma and BALF (P〈0.05). The pathological changes in lung tissue were lessened, and the expression of NF-κB p65 in lungs and W/D were decreased after penequinine hydrochloride injected (P〈0.05). Conclusion Penequinine hydrochloride has protective effects on paraquat-induced acute lung injury in rats.
