摘 要:为揭示鸡氨肽酶N(cAPN)含量差异与传染性支气管炎病毒(IBV)组织嗜性之间的关系,以看家基因鸡β-actin为内参基因,与cAPN目的基因在相同体系中扩增,以cAPN-pMD18-T标准质粒扩增曲线为标准,计算内参基因及目的基因各自的表达量,并将二者的比值作为相对定量依据,比较不同日龄雏鸡各组织cAPN含量的不同。结果显示,随着日龄增加,肝、脾、法氏囊、回肠、肾等组织中的cAPN表达量相对稳定,肺、腺胃、空肠等组织中的cAPN表达量呈现持续下降趋势,而十二指肠中的cAPN表达量持续上升,但盲肠、空肠中的cAPN表达量规律性不强且波动较大。表明cAPN的表达与IBV亲嗜组织存在着密切的相关性,同时也侧面反映了IBV组织嗜性的差异在一定程度上取决于机体不同组织器官cAPN的含量差异。To elucidate the relativity between chicken aminopeptidase N(cAPN) content diversity in different tissues and the tissue tropisms of infectious bronchitis virus(IBV) in chicks, genome RNA of heart, liver, spleen, lung, kindey, glandular stomach, pectoral muscle, duodenum, jejunum, ileum, appendix and bursa of Fabricius was extracted from chicks on different days of age,respectively,and the cAPN gene was amplified by RT-PCR. A 186 bp DNA fragment of cAPN was obtained by PCR and real time fluorescent quantitation PCR, then the expression level of cAPN mRNA was compared each other. The results showed that the expressed cAPN in different tissues displayed different change tendency with the increase of the age. The expression level of cAPN mRNA in lung,glandular stomach and jejunum were higher than that in other tissues,and the expression level of it in bursa of Fabricius and spleen were lower than that in other. The results also reflected that the tropism of IBV is closely related to the cAPN content diversity.
