摘 要:目的:观察芝麻素与维生素E联用对代谢综合征大鼠心肌损伤的保护作用。方法:高脂高糖诱导大鼠代谢综合征24周,第9周起每天口服1次芝麻素(30 mg/kg)、芝麻素+维生素E(30+20、15+20 mg/kg)和维生素E(20mg/kg)含药饲料,共16周。于末次给药后,称体重(BW)、全心(HWW)和左心室湿重(LVWW);测血糖、血脂和血压、心肌组织羟脯氨酸(HYP)、过氧化氢(H2O2)、一氧化氮(NO)含量及超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性;HE染色观察心肌组织病理变化;免疫组化法表达心肌诱导型一氧化氮合酶(iNOS)和硝基酪氨酸(NT)含量。结果:与模型组相比,芝麻素+维生素E(30+20 mg/kg)组血糖、血脂和血压明显降低,BW、HWW和LVWW显著下降;心肌H2O2、HYP和NO含量明显减少;SOD和CAT活性明显升高;心肌iNOS表达和NT含量减少;病理损伤明显减轻。上述结果优于单用芝麻素组或维生素E组(P<0.01或P<0.05)。结论:芝麻素(30mg/kg)与维生素E(20mg/kg)联用具有协同抗氧化和保护代谢综合征大鼠心肌的作用。Objective:To observe the synergistic protective effect of sesamin and vitamin E on myocardial damage in rats with metabolic syndrome.Methods: Metabolic syndrome was induced by high-fat and refined-carbohydrate diet for 24 weeks.Sesamin(30mg/kg),sesamin+vitamin E(15+20 mg/kg),sesamin +vitamin E(30+20 mg/kg) and vitamin E(20 mg/kg) were given to the metabolic syndrome rats at 9th week,which lasted for 16 weeks.Body weight(BW),heart wet weight(HWW)and left ventricle wet weight(LVWW) of all rats were weighted;blood glucose,blood lipids and systolic blood pressure were measured;the content of hydroxyproline(HYP),hydrogen dioxide(H2O2) and nitric oxide(NO),the activity of superoxide dismutase(SOD) and catalase(CAT) of myocardium were measured.Pathological change,the content of nitrotyrosine(NT) and expression of inducible nitric oxide synthase(iNOS) of myocardium were observed by H-E staining and immunohistochemical method,respectively.Results: Compared with model group,blood glucose,blood lipids,and blood pressure were decreased of sesamin +vitamin E(30 +20 mg/kg) group,the BW,HWW,LVWW,H2O2,HYP and NO were markedly reduced,and the activity of SOD and CAT obviously increased,the content of NT and expression of iNOS were relatively decreased,pathological change was markedly improved the therapeutic effect of sesamin +vitamin E(30 +20 mg/kg) surpassed the one of only using sesamin and vitamin E,respectively(P0.01or P0.05).Conclusion: Sesamin and Vitamin E have synergistic antioxidant activity and protective effect on myocardium of metabolic syndrome rats.
