摘 要:目的:观察丹酚酸B盐对阿霉素所致心肌损伤大鼠氧化应激反应和基质金属蛋白酶-2/9活性的影响。方法:除正常组外,SD大鼠尾静脉注射阿霉素复制心肌损伤模型。以不同浓度丹酚酸B盐灌胃,维生素E组灌胃维生素E乳浊液。HE染色观察心肌组织炎性变化,试剂盒方法测定血清乳酸脱氢酶活性、心肌组织总超氧化物歧化酶活性和丙二醛含量,明胶酶图法检测心肌组织基质金属蛋白酶-2/9活性。结果:与模型组相比,丹酚酸B盐能减轻心脏炎性病理,降低血清乳酸脱氢酶活性和心肌组织丙二醛水平,升高心肌组织超氧化物歧化酶活性,明显降低心肌组织MMP-2/9活性,以高、中剂量组作用更为显著。结论:丹酚酸B盐可以从改善心肌清除自由基能力和降低MMPs活性两方面,发挥对阿霉素所致心肌损伤的保护作用。Objective:To observe the effects of Salvianolic acid B(SA-B) on oxidative stress reaction and the activities of matrix metalloproteinases(MMP)-2/9 in the myocardium injury induced by adriamycin(ADR) in rat.Methods: SD rats were randomly divided into 6 groups:normal,model,SA-B high,SA-B middle,SA-B low and Vitamin E control group.Myocardium injury was induced by tail vein injection of ADR.The rats in SA-B groups were administered with deferent concentration of SA-B,and the rats in Vitmin E control group with Vitmain E from the day of first ADR injection.Myocardial inflammation were checked with HE staining,activity of lactate dehydrogenase(LDH),superoxide dismutase(SOD)and concentration of malondialdehyde(MDA) were assayed with kits.The activities of Myocardial tissue MMP-2/9 were determined with gelatin zymography.Results: SA-B can obviously attenuated heart inflammation,reduce the activity of LDH and MDA level,significantly decrease the activity of SOD and significantly down-regulated the activity of MMPs of myocardial tissues.The protection effects of SA-B high,middle groups were more significant.Conclusion: SA-B have protection effects on ADR induced myocardial injury,by improving the capability to clear oxygen free radical and decreasing the activities of MMP2/9.
