摘  要:目的探讨微小RNA(microRNA,miRNA)在子痫前期患者和正常产妇胎盘组织中的表达差异,及其在子痫发病中的作用。方法收集子痫前期患者和正常产妇胎盘组织并提取RNA;采用miRNA特异引物进行反转录;荧光实时定量聚合酶链反应(real-time PCR)检测miRNA-152(miR-152)和miR-377基因的表达量。结果在子痫前期患者胎盘组织中miR-152表达升高,miR-377表达降低。结论子痫前期患者胎盘组织中存在特异miRNA的差异表达,其发病可能受miRNA的调控。Objective: To detect the expression of microRNA-152 and microRNA-377 in human placentas of patients with preeclampsia, in order to investigate the role of small RNA (microRNA, miRNA) in the pathogenesis of preeclampsia. Methods: Placentas were obtained from patients with preeclampsia and normal parturients; Looped miRNA specific RT primers were used to reversely transcribe the miRNA, miRNA were then amplified by SYBR Green real-time RCR using miRNA specific PCR primers. Results: The expression of miRNA 152 in the human placentas of patients with preeclampsia was elevated, that of miRNA-377 was decreased. Conclusions: The present study suggests that the specific miRNA are distinctive expression in the human placentas of patients with preeclampsia, specific miRNA may involve in preeclampsia occurrence.
