摘  要:目的:研究黄芪对人乳腺癌细胞中MDA-MB-231系血管内皮生长因子(VEGF)及其受体(flk-1)和血小板源性因子受体-4(CXCR4)表达的影响。方法:在无酚红DMEM培养的人乳腺癌细胞中加0.5、1.0、2.0 g/L黄芪处理,对照组(Con)加等体积的蒸馏水。收集培养48 h的细胞,用免疫细胞化学和免疫印迹检测乳腺癌细胞中VEGF、flk-1和CXCR4蛋白的表达。结果:与Con组相比,黄芪组乳腺癌细胞中的VEGF、flk-1和CXCR4蛋白表达均下降,且高浓度组中表达明显降低。结论:高浓度黄芪能下调乳腺癌细胞中VEGF、flk-1和CXCR4蛋白的表达量,由此可能会减少肿瘤血管生成和降低肿瘤侵袭。Objective:To investigate the effects of astragalus mongholicus on the expressions of vascular endothelial growth factor(VEGF),VEGF receptor(flk-1),and CX chemokine receptor type 4(CXCR4) in MDA-MB-231 breast cancer cells.Methods:MDA-MB-231 breast cancer cells were cultured in phenol red-free DMEM and were treated with 0.5g/L,1.0g/L,2.0g/L astragalus mongholicus.The control group was treated with isovolumic distilled water.Western blotting and immunocytochemistry methods were used to detect the expressions of VEGF,CXCR4 and flk-1.Results:The expressions of VEGF,CXCR4 and flk-1 were lower than those of the control.There was significant difference between the high concentration group and the control.Conclusion:The expressions of VEGF,CXCR4 and flk-1 are down-regulated when breast cancer cells were treated with astragalus mongholicus at high concentration,therefore the angiogenesis and invasion of breast tumors maybe reduced.
