葛根素预处理减轻大鼠心肌缺血再灌注损伤及PI3K/AKT信号通路的作用  被引量:14

摘  要:目的:观察葛根素(Puerarin,Pue)预处理对缺血再灌注大鼠心肌的保护作用及其可能机制。方法:40只SD大鼠随机分为5组(n=8):假手术组(SH组);缺血再灌注组(I/R组);葛根素预处理组(PU组);葛根素预处理+LY294002组(PU+LY组);缺血再灌注+LY294002组(I/R+LY组)。结扎冠状动脉前降支30min,再灌注120min。于缺血前、缺血30min以及再灌注60、90、120min时,测定左室收缩压(Lef tventricular systolic pressure,LVSP)、左心室舒张末压(Left ventricular end-diastolic pressure,LVEDP)、左室内压变化最大上升和下降速率(Maximum rate of left ventricular pressure development and decay,±dp/dtmax);测定心肌梗死面积百分比、一氧化氮(Nitric oxide,NO)和一氧化氮合酶(Nitric-oxide synthase,NOS)的含量。结果:I/R组较SH组各时间点的收缩及舒张功能显著降低,心肌梗死面积增加,NO和NOS含量明显降低;与I/R组比较,PU组各时间点的收缩及舒张功能下降程度明显减轻,心肌梗死面积明显减少,NO、NOS含量明显增加;PU+LY组与PU组相比各时点的收缩及舒张功能显著降低,心肌梗死面积增加,NO和NOS含量明显降低;PU+LY组、I/R+LY组与I/R组相比以上指标差异均无显著意义。结论:葛根素预处理对缺血再灌注大鼠心肌有较好的保护作用,其作用机制可能与PI3K/Akt信号通路的活化有关。Objective: To investigate the protective effect of preconditioning with Puerarin on myocardial ischemia reperfusion in adult SD rats and its possible mechanism. Methods: Forty Male SD rats were divided randomly into five groups (n=8). The myocardial injury of rats ware produced by the ligation of left coronary artery for 30 min followed by 120 min reperfusion. LVSP, LVEDP, and ± dp/dtmax were recorded at ischemia 30 min, reperfusion 60, 90 and 120 min. The myocardial infarct size and the level of NO and NOS were observed. Results: In group I/R, the systolic and diastolic function were significantly reduced ; myocardial infarct size was increased ; NO and NOS were significantly lowered compared with those in group SH(P〈0.05 ).The systolic and diastolic function were improved;myocardial infarct size was significantly smaller; the level of NO and NOS were increased significantly in group PU as compared with those in group I/R(P〈0.05 ). All these parameters in group PU+LY were not improved compared with those in group PU(P〈0.05 ). There was no significant difference among the parameters of groups PU+LY, I/R+LY,and I/R. Conclusion: Preconditioning with puerarin can significantly reduce myocardial ischemia-reperfusion injury, the mechanism of which is partly via PI3K-Akt signal transduction.
