摘  要:目的探讨仿真手针预处理对内脏牵拉痛大鼠的行为学的影响及其抗大鼠内脏牵拉痛的作用机制。方法40只雄性SD大鼠随机分4组(假手术组、VTP组、仿真手针组、电针组),每组10只;后2组分别进行针刺、电针30min后即刻制备VTP模型,观察各组大鼠的疼痛积分,并分别采用免疫组化、生化比色法、放射免疫等方法检测大鼠回肠部肠神经系统的SP阳性表达物、AChE的活性和LEK的含量以及延髓内脏带c-Fos蛋白和GFAP阳性指数的含量。结果与假手术组比较,VTP组大鼠疼痛积分显著升高(P<0.05),回肠部SP阳性表达物、AChE的活性,延髓内脏带c-Fos蛋白、GFAP阳性指数的含量显著增高(均P<0.01),但LEK含量增加不显著(P>0.05);与VTP组比较,仿真手针和电针组大鼠疼痛积分,回肠部SP阳性表达物、AChE的活性,延髓内脏带c-Fos蛋白、GFAP阳性指数的含量显著下降(均P<0.05),电针组LEK含量增加不显著(P>0.05),仿真手针组LEK含量明显增加(P<0.05);与电针组比较,仿真手针组除疼痛积分显著降低(P<0.05)和LEK含量显著增高(P<0.05)外,其他指标无显著性差异(均P>0.05)。结论仿真手针预处理能通过减少SP的释放,减低AChE的活性,增加LEK的释放,下调c-Fos蛋白、GFAP的表达,有效抑制内脏牵拉痛,而仿真手针预处理抑制内脏牵拉痛的效果优于电针,其原因在于仿真手针更能有效增加LEK的释放。Objective To investigate the effect of emulation acupuncture pretreatment on the behavior of rats with visceral traction pain and the mechanism of its relieving action on rat visceral traction pain. Methods Forty male SD rats were randomly allocated to four groups (sham operation, VTP, emulation acupuncture and electroacupuncture), 10 rats each. A VTP model was made shortly after 30 min treatment. The pain score was counted in each group of rats. Rat ileal nerve SP expression-positive substance, AChE activity and LEK content and bulbar visceral zone c-Fos protein and GFAP positive indices were examined using an immunohistochemical method, biochemical colorimetry and radioimmunoassay. Results The pain score (P〈0.05), ileal SP expression-positive substance and AChE activity and bulbar visceral zone c-Fos protein and GFAP positive indices (all P〈0.01) increased significantly but LEK content (P〉0.05) did not in the VTP group compared with the sham operation group. The pain score, ileal SP expression-positive substance and AChE activity and bulbar visceral zone c-Fos protein and GFAP positive indices decreased significantly in the emulation acupuncture and electroacupuncture groups (all P〈0.05) and LEK content did not increased obviously in the electroacupuncture group (P〈0.05) but did in the emulation acupuncture group (P〈0.05) as compared with the VTP group. The pain score decreased significantly (P〈0.05), LEK content increased significantly (P〈0.05) and the other indices did not change significantly (all P〉0.05) in the emulation acupuncture group compared with the electroacupuncture group. Conclusion Emulation acupuncture pretreatment can decrease AChE activity, increase LEK release, down-regulate c-Fos protein and GFAP expressions and effectively relieve visceral traction pain through reducing SP release.
