摘 要:[目的]分析福建省流行的肠道病毒71(Enterovirus71,EV 71)分离株的遗传特征。[方法]对2008年手足口病例(HFMD)EV 71分离株进行RT-PCR扩增全长VP1基因,测序并分析病毒遗传学特征。[结果]从福州、泉州及龙岩3地市散发的手足口病例中分离到8株EV 71,种系发生树显示2008年福建与阜阳分离株和中国大陆1998年以来的病毒株均为C4亚型,至少分为4个谱系(lineages)。8株福建分离株核苷酸与氨基酸同源性分别为97.4%-100%和99.3%-100%,与Genbank安徽阜阳的5株分离株病毒同源性最高,核苷酸与氨基酸序列同源性为97.6%-99.5%和96.4%-99.3%,其中福建株HF08-17、HS08-140与阜阳分离株同源性为99.2%-99.5%,为同一传播链上的病毒;福清市2分离株(HA08-68和HA08-69)以及长汀县3株(HA08-124,HS08-139和HS08-140)病毒间核苷酸差异性为2.1%-2.2%,说明同一地区存在不同传播链。[结论]2008年福建省EV 71分离株均属于C4亚型,C4亚型病毒在中国大陆具有多传播链、广泛快速传播的特点;福建省内存在亲缘关系较近的EV71多传播链病毒同时流行,应加强监测与分析。[Objective] To analyze the genetic characteristics of enterovirus 71 (EV 71) strains isolated from suspected patients of hand foot-and-mouth disease (HFMD) in Fujian,2008. EMethods2 The VP1 genes of EV 71 strains were amplified by RT PCR and sequences were analyzed by software. [Results] Eight EV 71 strains were isolated from the sporadic HFMD cases of Fuzhou,Quanzhou and Longyan in 2008. All strains from Fujian,Fuyang,and other reference strains from China's Mainland after 1998 belonged to C4 subgenotype,and could be divided into at least 4 lineages, according to the phylogenetic tree. Within the Fujian isolates, the nucleotide and amino acid identity was 97.4%-100% and 99.3%-100% respectively,and Fujian stains shared high homology to Fuyang isolates available at Genbank,with 97.6%-99.5% nucleotide identity and 96. 4%- 99. 3% amino acid identity. Two isolates from Fujian, HF08-17 and HS08 140,were 99.2%-99.5% identical to Fuyang isolates, suggested that they were in the same transmission chain. The nueleotide difference between two strains isolated in Fuqing and three isolated from Changting was 2. 1%-2.2%, suggested there were more than one transmission chain in a county. [Conclusions] EV 71 strains in Fujian isolated in 2008 were of C4 subgenotype. C4 strains in mainland of China were characterized with multiple transmission chains and quick transmission. Extensive surveillance and analysis are required to trace the co-circulation of multiple transmission chains caused by closely'related EV 71 strains in Fujian.
