摘  要:目的测定3-氧-乙酰-11-脱氧甘草次酸铝(Aluminum3-o-acetyl-11-deoxoglycyrh-ate,ADA)的半数致死量(LD50)及对大鼠血清钠、钾浓度的影响。方法昆明小鼠60只,体重20±1g,随机分6组,每组10只,各组按不同剂量的ADA0.2ml·10g-1灌胃,给药后按急性毒性实验法连续观察7d。Wistar大鼠40只,体重200±10g,随机分4组,每组10只。各组分别给0.5%羧甲基纤维素钠(CMC)、生胃酮(Carbenoxolone,CN)和两个剂量的ADA,早晚灌胃一次,连续12d后,处死大鼠取血,测血清钠、钾浓度。结果灌胃ADA的LD50为4444±831.5mg·kg-1。长期用药,ADA未引起大鼠血清钠、钾浓度的明显变化,相反CN却显著降低血清钾浓度。结论ADA引起急性致死性中毒的危险性甚小,长期用药未引起大鼠血清钠。Objective To measure the median lethal dose of aluminium 3-o-acetyl-11-deoxoglycyrrhate(ADA) and the effect on Na+ and K+ serum concentration in rats.Methods The 60 mice weighing 20±1g,were randomly divided into 6 groups which given orally different doses of ADA 0.2ml·10g^-1 respectively. Then the observation lasted 7days according to the acute toxic method. The 40 Wistar rats weighing 200 ±10g, were randomlydivided into 4 groups which were given 0.5% sodium cellothyll carboxymethylcellulose(CMC),carbenoxolone(CN) and two doses of ADA respectively. The route was given ig,twice a day,for 12 days.Then rats were killed and the blood was collected to measure Na^+ and K^+ concentrations in serum.Results The ig median lethal dose of ADA is 4444±831.5mg·kg^-1.Serum Na^+ and K^+ concentrations are not affected by ADA after the drug was given orally for consecutive 12 days. But CN significantly decrease the K^+ concentration.
