混合胶束扫集毛细管电泳法分离补骨脂中有效成分  

Simultaneous Seperation of Psoralen and Isopsoraden in Psoralea Corylifolia L. by Sweeping Micellar Electrokinetic Capillary Chromatography

摘　　要：采用混合胶束毛细管电泳在线扫集技术分离测定了中药补骨脂中补骨脂素和异补骨脂素。电泳条件:以35mmol/L硼砂-40mmol/LSDS-5mmol/Lβ-环糊精(CD)-5mmol/L尿素(含10%体积分数乙醇,pH2.8)为电泳运行缓冲溶液,未涂层石英毛细管(φ50μm×58cm,有效长度为41.2cm)为分离通道,重力进样,进样高度为11cm,-20kV恒压,检测波长为246nm。通过此方法扫集,富集倍数可以达到160倍以上,极大地提高了检测的灵敏度。A sweeping micellar eletrokinetic capillary chromatographic method has been developed for separation and quantitative analysis of Psoralen and Isopsoraden in Psoralea corylifolia.Electroretic conditions are as follows:uncolated fused silica capillary(41.2 cm/58 cm,50μm),35 mmol/L disodium tetraborate decahydrate,40 mmol/L SDS containing 10% ethanol(V/V)at pH 2.8,-20 kV applied voltage,detection wave-length 246 nm,injection by gravity with the height 11cm.Up to 160-fold the improvement in multiple of enrichment is achieved by using this sweeping injection technique.
