摘  要:目的:探讨2-甲氧雌二醇(2-ME)对HL60白血病细胞增殖及凋亡的影响。方法:用MTT法和流式细胞术检测2-ME对HL60的增殖抑制及促凋亡作用,用流式细胞技术检测Bcl-2蛋白的表达。结果:随着2-ME浓度升高及作用时间延长,HL60细胞增殖明显受抑制(P<0.05),细胞凋亡率明显增加(P<0.05),当2-ME浓度为50μmol/L时,细胞凋亡率达65.8%;2-ME(30μmol/L)作用72h后,Bcl-2的表达较对照组显著减少(P<0.05)。结论:2-ME对HL60细胞增殖有抑制及促凋亡作用,Bcl-2表达下调在此过程中可能发挥重要作用。Objective:To investigate the effect of 2-Methoxyestradiol(2-ME)on the proliferation and apoptosis of HL60 cells in vitro. Methods:After treatment with 2-ME,the cell proliferation was detected with MTT,and the apoptosis was determined with flow cytometry. Expression of Bcl-2 protein in HL60 cell was detected with flow cytometry. Results:The growth inhibitation rate exhibited significant increases(P〈0.05)with the increased dose and extended time. The cells apoptotic rate increased significantly after treatment with 2-ME respectively(P〈0.05). The cells apoptotic rate was 65.8% after treatment with 2-ME(50 μmol/L)for 72 hours. Bcl-2 decreased significantly compared with control group after treatment with 2-ME(30 μmol/L)for 72 hours(P〈0.05). Conclusion:2-ME can inhibit proliferation and induce apoptosis of HL60 in vitro. The down regulation of Bcl-2 may play an important role in the procedure.
