摘 要:目的研究黄芪苷Ⅳ(XGA)对损伤的人血白细胞变形能力的影响。方法人血白细胞经孵化后损伤,用核孔膜滤过法观察XGA对白细胞变形能力的影响,并在电镜下观察白细胞形态的变化。结果人血白细胞在孵化24及48h后受到损伤,白细胞阻塞率(CI)值明显增加,50μg/ml样品的XGA对损伤的白细胞能明显降低其CI值,改善白细胞的变形能力;XGA在25~250μg/ml样品的剂量范围内对损伤的白细胞均能明显降低其CI值,改善白细胞的变形能力;电镜下观察到50μg/ml样品的XGA对损伤的白细胞有修复作用。结论XGA能明显改善受损白细胞的变形能力,而白细胞变形能力的改善可能是XGA防治心脑血管疾病的作用机制之一。PURPOSE To study the influence of astragaloside Ⅵ(XGA) on injured human blood leukocyte deformability. METHODS The human blood leukocyte was injured after incubation. Nuclepore filtration technique was applied to observe the influence of XGA on injured human blood leukocyte deformability. The morphological changes of leukocyte were observated under electron microscopy. RESULTS The human blood leukocyte was injured after incubation for 24 and 48 hours, and CI was significantly increased. The CI of the injured leukocyte was significantly decreased when XGA was given at a concentration of 50μg/ml solution, and the deformability of leukocyte was improved. When XGA was given at a concentration of 25~250μg/ml, the CI of the injured leukocyte was significantly decreased and the deformability was obviously improve. Electron microscopic observation indicated that XGA possessed raparation effect on injured leukocyte. CONCLUSIONS XGA can obviously improve the deformability of leukocyte. This improvement is possibly one of its mechanisms in preventing and treating cardiovascular diseases.
