摘  要:目的研究CSF1PO、TH01和TPOX3个短串联重复序列(STR)基因座的等位基因频率在洛阳地区汉族群体内的分布,为这3个基因座的应用提供基础数据。方法用Chelex100法提取抗凝血的DNA,用PowerPlex(16系统扩增样品DNA,扩增产物经ABIPRISM(377DNA Sequencer电泳后,用genescan3.1和genotyper2.1软件确定各等位基因。统计各等位基因频率,用2检验检验遗传平衡吻合度。计算杂合度(H)、多态信息含量(PIC)、个人识别概率(DP)及亲权否定概率(PPE)。结果得到3个基因座各等位基因及其频率,其基因型分布符合遗传平衡定律,具有高的杂合度和多态信息含量。结论3个基因座具有较高的杂合度,是较理想的遗传标记,可用于法医学个人识别和亲权鉴定以及人类遗传学研究。Objective To investigate the allele frequencies of CSF1PO,TH01 and TPOX in Luoyang Han population and to provide basic data for their use in related field.Methods DNA extracted with Chelex100 from anti-coagulative blood sample was amplified with PowerPlex 16 system.The PCR product was analyzed with ABI PRISM 377 DNA Sequencer and typed with genescan2.1 and genotyper3.1 software.After getting the allele frequencies and phenotype frequencies,Χ^2 test was performed to determine if it followed Hardy-Weinberg' s law. Based on the allele frequencies, the value of heterozygosity (H),polymorphism information content ( PIC), discrimination probability- (DP) and exclusion power ( PE ) were calculated according to related formula. Results Allele frequencies were gotten and the allele distribution agreed with Hardy-Weinberg equilibrium. The genetic parameters, such as heterozygosity(H) , polymorphism information content(PIC) , power of discrimination(DP) , probability of paternity exclusion(PPE) , were also calculated. Conclusion These three STR loci possess high heterozygosity and can be used as genetic markers in human genetic study, personal identification and paternity test.
