饮食对单次口服乐福昔布后生物利用度的影响

摘　　要：目的研究Beagle犬禁食和非禁食情况下单次口服乐福昔布(25.5mg.kg-1)片剂后药动学参数变化规律以及绝对生物利用度。方法以塞来昔布为内标,经方法学验证后的HPLC-荧光分析方法,测定给药后Beagle犬各时间点血浆中乐福昔布的浓度。结果Beagle犬在禁食和非禁食情况下乐福昔布片剂在Beagle犬体内的绝对生物利用度分别为3.8%和29.7%。结论饮食对乐福昔布在体内的吸收有显著影响。Aim To study the difference in pharmacokinetics and relative bioavailability of lefucoxib between Beagle dogs＇ intake of food and fasting before being given lefucoxib.Methods Celecoxib was used as the internal standard.Lefucoxib in Beagle dog plasma was determined by high-performance liquid chromatography with fluorescence detection.Results The lefucoxib＇ relative bioavailability was 3.98% and 29.7% respectively in case of intake of food and fasting.Conclusion Food can improve organisms＇ absorption of lefucoxib in vivo.
