摘 要:目的探讨前列腺素 E_1(PGE_1)对大鼠实验性胰腺炎的影响。方法向大鼠胰管内注入5%牛磺胆酸钠,制成急性坏死性胰腺炎模型。结果胰腺炎时胰头部血流明显下降,胰腺毛细血管通透性明显升高。胰腺组织中性粒细胞髓过氧化物酶(MPO)、脂质过氧化物(LPO)及血浆一氧化氮(NO)明显升高。病理检查示胰腺水肿、中性粒细胞(PMN)浸润、出血及坏死。预先使用 PGE_1,可明显增加胰头部血流,降低胰腺毛细血管通透性和胰腺 MPO、LPO 及血浆 NO 含量。胰腺以水肿为主,出血及坏死明显减轻,动物存活明显改善。结论 PGE_1 可通过抑制 PMN 活化及抑制氧自由基释放,减轻内皮细胞损伤,增加胰腺血流量,并减轻胰腺出血和坏死,改善存活质量。PGE_1抑制 NO,对于减缓低血压,提高胰腺血流量可能具有重要意义。To investigate the effect of prostaglandin E_1(PGE_1) on the course of experimental pancreatitis of rats.Methods The model of acute necrotizing pancreatitis (ANP) was induced by retrograde infusion of 5% sodium taurocholate into the pancreatic duct of rats.Results It was showed that blood flow in pancreatic head (BFPH) de- creased,while pancreatic capillary permeability(PCP),the activity of myeloperoxidase(MPO) and levels of lipoperox- ides(LPO) in pancreas and nitric oxide(NO) in plasma signilicantly increased during ANP.The pretreatment by PGE_1 anticipated or ameliorated the abovementioned adverse pathophysiological changes seen in control.Conclusion PGE_1 raised BFPH,prevented pancreatic bleeding and necrosis,improved survival of rats by means of inhibiting PMN acti- vation and release of OFR.
