摘 要:目的探讨镁离子、山药多糖单独应用和联合应对四氯化碳肝损伤的保护作用。方法50只小鼠随机分为5组,分正常对照组、病理造模组、山药多糖保护组(按100mg/kg体重山药多糖灌胃)、镁离子保护组(饮含20mmol/L硫酸镁的自来水)、镁离子山药多糖保护组(饮含20mmol/L硫酸镁的自来水,按100mg/kg体重山药多糖灌胃)、饲养8d,末次灌胃2h,正常对照组腹腔注射调和油溶液,其余各组腹腔注射0.15%四氯化碳调和油溶液,12h后禁食不禁水,24h眼球取血,分离血清测定ALT、AST,MDA。取出肝脏称重计算肝体指数,用pH7.4的PBS制备10%肝匀浆,测定MDA、GSH、SOD、NOS及NO含量。结果镁离子、山药多糖保护组能降低肝体指数及血清ALT和AST,与模型组比较差异有显著性(P<0.05~0.01)。提高GSH含量,降低MDA、NO含量和NOS活性,镁离子和山药多糖联合应用保护作用加强,与模型组比较(P<0.01~0.001)。结论镁离子、山药多糖对四氯化碳肝损伤有保护作用,联合应用有协同效应。[ Objective ] To investigate the protective effects of Mg^2+and yam polysaccharide on CCL4 liver injury when individual and joint application. [ Method ] 50 mice were randomly divided into five groups: Control group, Pathological model group, Yam polysaceharide protection group in which experimental mice were intra-gastrically treated according to 100 mg/kg body weight, Mg^2+ protection group in which experimental mice were intra-gastrieally done with 20 mmol/L magnesium sulfate, and Yam polysaccharide plus Mg^2+ protection group in which experimental mice were intra-gastrically done with 20 mmol/L magnesium sulfate and 100 mg/kg weight of yam polysaccharide. After the experimental mice were feed for seven days according their planned diets, Control group was intra-peritoneaUy injected the mixed solution, the other groups were intra-peritoneally injected the mixed solution with 0.15% CCL4. 12 hours later, the subjective were only given water, not given any food and keeping for 12 hours, the blood samples were prepared from mice's eyeballs for the determination of serum ALT, AST, MDA, the livers were weighted and 10% liver homogenate were done with PBS for the examination of MDA, GSH, SOD, NOS and NO contents. [Results] Compared with Control group (P 〈0.05-0.01), Mg^2+ and yam polysaccharide protection groups reduced the liver index, serum ALT and AST levels, lower MDA, NO content and NOS activity and improve GSH content. [ Conclusion ] Mg^2+ or yam polysaccharide has the protective effect on CCL4-indueed liver injury. Furthermore, the combined application of Mg^2+ with yam polysaccharide has a synergistic effect.
