莪术油对大鼠乳腺癌癌前病变组织中Ki67表达的影响

摘　　要：目的:通过观察莪术油对大鼠乳腺癌癌前病变组织中Ki67的表达,探讨莪术油治疗乳腺癌前病变的机理。方法:275只SD大鼠随机分为空白对照组、疾病模型组、三苯氧胺组、康莱特组及莪术油小、中、大剂量组。采用DMBA诱导乳腺癌癌前病变造模,干预治疗4周,第8～14周分4批处死动物,进行病理学及Ki67免疫组化检测。结果:各试验组不同类型乳腺组织中Ki67表达阳性率及阳性细胞阳性率呈递增趋势。非典型增生乳腺组织中,各干预组Ki67表达阳性率均明显低于造模对照组(P<0.05)。各干预组的Ki67阳性细胞阳性率均明显低于造模对照组(P<0.05),莪术油组低于康莱特和三苯氧胺组(P<0.05)。对于Ki67表达阳性的非典型增生标本,莪术油大剂量组Ki67阳性细胞阳性率明显低于小剂量组(P<0.05)。结论:莪术油能够降低DMBA诱导大鼠乳腺癌癌前病变组织中上皮细胞的增殖活性,可能是其阻断乳腺癌发生的有效机制。Objective：To research the treatment mechanism of Zedoray Turmeric Oil（ZTO） in precancerous lesion of rats mammary by measuring the expression of Ki67. Methods ： 275 SD rats were randomly divided into 7 groups ： the blank control group（ BC）, the precancerous lesion model control group（ PL）, the kanglalte control group （KLT） , the tamoxifen control group （TAM） , the low dose group of ZTO （ZTO -L） , the middle dose group of ZTO （ZTO -M） and the high dose group of ZTO （ZTO -H）. Animal model of precancerous lesion of rats mammary was induced bye DMBA and was treated by ZTO, KLT and TAM for 4 weeks. All the animals were killed from 8th to 14th week once every two weeks. Examination of histopathology was performed and measured the expression of Ki67 by immunohistochemistry. Results ： The positive rate of expression of Ki67 and the positive - cell rate trend to increase in every groups except BC corresponding to normal, usual ductal hyperplasia（ UDH）, atypical ductal hyperplasia（ADH） and invasive carcinoma. In ADH, the posi tive rate of expression of Ki67 of ZTO, KLT and TAM was much lower than PL（P 〈0.05）. The positive -cell rate of Ki67 of ZTO, KLT and TAM was much lower than PL（ P 〈 0.05）, and the positivecell rate of Ki67 of ZTO was much lower than KLT and TAM （P 〈 0.05 ）. In the case of ADH which the expression of Ki67 was positive, the positivecell rate of Ki67 of ZTO - H was was much lower than ZTO - L（P 〈0.05）. Conclusion： Zedoray Turmeric Oil decrease the proliferation of breast epithelial tissue in precancerous lesion of rats mammary induced by DMBA, which could be the mechanism to prevent the breast cancer.
