摘 要:目的:观察槟榔对离体大鼠结肠平滑肌收缩运动的影响,并初步探讨其机制。方法:制备大鼠结肠平滑肌肌条,以9g/LNaCl溶液(NS)、槟榔为对照组,分别记录单独使用槟榔和阻断剂(酚妥拉明、维拉帕米和阿托品)孵育后槟榔引起的收缩效应。结果:不同浓度槟榔对离体大鼠结肠平滑肌的促进作用明显,与NS对照组比较,槟榔0.1g/L、1g/L、10g/L浓度时引起的肌条收缩最大振幅和面积均有统计学意义(P<0.05);与单独槟榔组比较,阻断剂维拉帕米和阿托品对槟榔引起的肌条收缩有抑制作用,其最大收缩振幅和面积均有统计学意义(P<0.05);而阻断剂酚妥拉明对肌条的收缩振幅和面积的影响均无统计学意义(P>0.05)。结论:槟榔对大鼠结肠平滑肌的收缩活动有兴奋作用,且呈正相剂量关系,引起的收缩效应可被M受体阻断剂阿托品和Ca2+阻断剂维拉帕米阻断,而未能被α-受体阻断剂酚妥拉明阻断。Objective :To determine the effect of Areca Seed (AS) on murine colonic smooth muscle strips in vitro and to investigate its possible mechanism. Methods :The colonic longitudinal smooth muscle strips of rats were prepared. Control group was treated with sodium chloride (9g/L) and AS. We observed the effects of AS on the spontaneous contraction in muscle strips. We also observed its promotion effects on muscle strips fully incubated with Verapamil, Phentolamine and Atropine, respectively. Results : AS of different concentrations showed dose - dependent promotion effects on the spontaneous contraction of colonic muscle strips in vitro at the concentration of 1 - 1000g/L. Compared with NS group, the final concentration of AS 0. 1g/L, 1g/L and 10g/L, the peak value and area data of smooth muscle strips have significant difference (P 〈 0.05). Compared with AS group, the peak value and area data of smooth muscle strips were inhibited in Atropine and Verapami group was inhibited significantly (P 〈 0.05 ) , but there was no significant difference of the peak and area in Phentolarnine group. Conclusion : AS showed promotion effects on spontaneous contraction of murine colonic smooth muscle strips in vitro. Its possible inhibitory mechanism has close relations with the calcium and perhaps with the M - cholinergie receptor, but has little relation with the α - adrenoceptor receptor.
