摘 要:目的:探讨人胎盘提取液对高脂饮食性脂肪肝的治疗作用及对血清及肝组织中Leptin的影响。方法:42只Wistar大鼠,雄性,分为正常组、模型组、易善复组、胎盘提取液高剂量组和低剂量组,予既定试剂4周后,取肝组织行HE染色观察组织学改变,用放免法检测各组大鼠血清及肝组织中Leptin的含量。结果:组织学观察表明造模组均出现了中度以上大泡性肝脂肪变性,人胎盘提取液低剂量组和易善复组较造模组有好转,但无明显差异(P>0.05),人胎盘提取液高剂量组脂肪浸润状态较造模组有明显好转,且差异明显(P<0.05)。造模组大鼠血清、肝组织中Leptin的含量均高于正常对照组(P<0.05或P<0.01〉,人胎盘提取液高、低剂量组、易善复对照组较造模组明显降低(P<0.05或P<0.01)。结论:细胞因子Leptin参与脂肪性肝病的发生。人胎盘对高脂饮食脂肪肝具有明显的治疗作用,对细胞因子Leptin具有明显的抑制作用。Objective: To investigate the effects of human placenta to the rats with nonalcoholic fatty liver disease and its influence on the cytokines such as Leptin. Methods: Forty - two male Wistar rats were randomly divided into four groups :normal group, model group, Essentiale group , human placenta high dose group and human placenta low dose group. After treated with different agentia already definited for four weeks,histological examinations of liver were carried out with HE staining; meanwhile, the serum and tissue levels of Leptin were detected with RIA. Results:Histological examinations showed that there were severe fatty degeneration of hepatocytes in the model group. The damage in livers with low dose human placenta extracts and Essentiale was shghtly milder than that of the models(P 〉0.05). While high dose human placenta could obviously lessen the fatty degeneration ( P 〈 0.05 ). Compared with the blank control group, the serum and tissue levels of Leptin in the model group were all remarkably increased( P 〈0.05 or P 〈0.01 ). And the human placenta and Essentiale could significandy lower the levels of Leptin (P 〈 0.05 or P 〈 0.01 ). Conclusion:Human placen- ta is effective in treating fatty liver induced by high - fatty diet. This may be due to its effects in lowering the levels of the Leptin which were concerned with the pathogenesis of nonalcoholic fatty liver disease.
