摘  要:目的:观察桑精胶囊对2型糖尿病大鼠脂肪和骨骼肌组织胰岛素受体底物1(insulin receptor substrat1,IRS-1)蛋白表达及其丝氨酸磷酸化水平的影响,探讨其治疗2型糖尿病的分子机制。方法:采用尾静脉注射小剂量链脲佐菌素(strepto-zotocin,STZ)加高脂高热卡饲料喂养的方法建立2型糖尿病(type2diabetes mellitus,T2DM)大鼠模型,将造模动物随机分为模型组、二甲双胍组和桑精胶囊组,另设正常对照组。药物干预8周后,用免疫沉淀和Western印迹方法检测脂肪和骨骼肌组织胰岛素刺激前和胰岛素刺激后IRS-1表达水平和丝氨酸磷酸化水平。结果:与模型组比较,桑精胶囊组胰岛素刺激前和胰岛素刺激后脂肪和骨骼肌组织中IRS-1蛋白表达均增高,IRS-1丝氨酸磷酸化水平均显著下降(P<0.05或P<0.01)。结论:桑精胶囊对2型糖尿病的治疗效应可能与提高脂肪和骨骼肌组织中IRS-1蛋白表达并抑制其丝氨酸磷酸化水平有关。OBJECTIVE To investigate the molecular mechanism of Sangjing capsule (SJC) in treating murine type 2 diabetes mellitus(T2DM) by observing its effects on insulin receptor substrate 1 (IRS-l)protein expression and serine phosphorylation in adipose and skeletal muscle tissues. METHODS T2DM in rats was induced by intravenous injection of a small dose (25 mg·kg^-1) of streptozotocin plus high fat and high caloric laboratory chow. Then animals were divided into untreated group, metformin treated group and SJC treated group. The normal rats fed with common chow were designated as normal control group. Eight weeks later the expression of IRS-1 protein and serine phosphorylation in adipose tissues and skeletal muscle were detected before and after insulin stimulation were measured. RESULTS Compared with the untreated group, the results of the levels of IRS-1 serine phosphorylation (Ser307) of the SJC group after treatment were decreased while the IRS-1 protein expression was elevated obviously (P〈00. 05 or P〈0. 01). CONCLUSION It is suggested that the therapeutic effects of SJC on T2DM might be related to its ability of increasing IRSyl protein expression and reduced IRS-1 protein serine phosphorylation in adipose and skeletal muscle tissues.
