摘 要:目的:探讨硼替佐米对卵巢癌SKOV-3/DDP细胞增殖和耐药逆转的影响和作用机制。方法:采用MTT法检测不同浓度硼替佐米和顺铂(DDP,cisplatin)对SKOV-3/DDP不同作用时间的细胞生长抑制率,以及硼替佐米和DDP合用和单独应用时的细胞生长抑制率;流式细胞术检测细胞凋亡和细胞周期的变化。结果:硼替佐米能够有效抑制SKOV-3/DDP细胞增殖,硼替佐米与DDP合用组的SKOV-3/DDP细胞抑制率较硼替佐米和DDP单用组均明显增强。单独应用硼替佐米细胞周期阻滞于G2/M期,单独应用DDP细胞周期阻滞于S期,两药联合出现更明显的S期阻滞。结论:硼替佐米与DDP具有协同作用,能够逆转SKOV-3/DDP细胞对DDP的耐药性。OBJECTIVE To investigate the effect and mechanism of bortezomib inhibiting cell growth and reversing drug resistance of ovarian cancer cell line SKOV 3/DDP in vitro. METHODS SKOV-3/DDP cells were treated by different eoncentra tions of bortezomib and DDP respectively at different time intervals, then the growing inhibiting rates were examined with MTT; and the cellular apoptosis percentage and cell cycle phase distribution were studied by flow cytometry (FCM). RESULTS Bortezomib had great inhibitory effect on the growth of SKOV- 3/DDP cell. The growth apoptosis rate of bortezomib combined with DDP was more obvious than either one used separately. The cell cycle was blocked at G2/M stage when using bortezomib singlely, which was blocked at S stage when using DDP singlely, while the cell cycle was blocked at S stage more obviously when using both of them. CONCLUSION Bortezomib and DDP have synergistic effect, they can reverse the drug re sistance of SKOV-3/DDP cell to DDP.
