摘  要:目的:观察阿托伐他汀对兔脂肪细胞型脂肪酸结合蛋白(AFABP)表达的影响。方法:取新西兰兔皮下脂肪组织行脂肪细胞原代培养,分别给予不同浓度的氧化型低密度脂蛋白(OxLDL)及阿托伐他汀干预。采用酶联免疫吸附法检测脂肪细胞培养液中AFABP水平,半定量逆转录多聚酶链式反应测定脂肪细胞AFABP和PPARγmRNA的表达。结果:较低剂量的OxLDL呈剂量依赖性刺激脂肪细胞AFABP表达和分泌,并上调脂肪细胞PPARγmRNA的表达;阿托伐他汀呈剂量依赖性降低OxLDL诱导的脂肪细胞AFABP分泌及mRNA表达。结论:阿托伐他汀可显著抑制OxLDL诱导的兔脂肪细胞AFABP的表达和分泌,这一作用可能有利于动脉粥样硬化的防治。OBJECTIVE To evaluate the effects of atorvastatin on adipocyte fatty acid binding protein(AFABP)secretion and expression induced by oxidized low density lipoprotein (OxLDL) in adipocytes. METHODS Subcutaneous adipose tissues were collected from normal rabbits for adipocytes culture. The adipocytes were exposed to OxLDL and atorvastatin at different concentrations. AFABP concentrations in adipocytes culture supernatant were measured by ELISA. The levels of AFABP and PPARγ mRNA in adipocytes were measured by semi-quantitative reverse transcriptiompolymerase chain reaction (RT-PCR) studies. RESULTS The OxLDL in low concentrations stimulated adipocytes release of AFABP; furthermore, OxLDL increased AFABP and PPARγ mRNA expressions of adipocytes in a dose-dependent manner. The treatment of adipocytes with atorvastatin inhibited AFABP mRNA expression induced by OxLDL and reduced release of AFABP in adipocytes with a dosedependent manner. CONCLUSION These findings indicated that atorvastatin may inhibit AFABP expression and secretion in duced by OxLDL in adipocytes which might be of significance for the prevention and treatment of atherosclerosis.
