摘  要:目的探讨沙苑子黄酮(FAC)对D-半乳糖(D-gal)致衰老模型小鼠的延缓衰老作用及其机制。方法选择ICR小鼠60只,将其随机分为6组:正常对照组(NG组)、模型对照组(MG组)、阳性药对照组(VE组,25mg·kg-1.d-1)、FAC高剂量组(FAC-H组,200mg·kg-1.d-1)、FAC中剂量组(FAC-M组,100mg.kg-1.d-1)及FAC低剂量组(FAC-L组,50mg·kg-1.d-1),各10只。除NG组外,其余5组使用D-gal按120mg·kg-1.d-1于小鼠背部皮下注射,连续30d,造成亚急性衰老模型。从造模的第21天开始,VE组小鼠灌服25mg.kg-1.d-1维生素,MG组小鼠灌服等体积0.9%氯化钠注射液,FAC-H、FAC-M、FAC-L组小鼠分别灌服相应浓度的FAC,连续10d。末次给药后30min从眼眶取血,按照测试盒要求检测并比较6组小鼠的自由基代谢〔超氧化物歧化酶(SOD)活性及丙二醛(MDA)、一氧化氮(NO)水平〕及免疫力指标〔胸腺指数(SI)、脾脏指数(TI)、碳廓清指数(K)及淋巴细胞转化率(用570nm测得OD值A代替)〕。本实验重复进行3次,所得数据取平均值。结果(1)与NG组比较,MG组小鼠血清SOD活性降低,MDA、NO水平升高,差异均有统计学意义(P<0.01)。FAC-H组、FAC-M组、FAC-L组小鼠血清SOD活性较MG组显著提高,血清MDA、NO水平较MG组显著下降,差异均有统计学意义(P<0.01)。(2)MG组小鼠的TI、SI、K、A与NG组比较,差异均有统计学意义(P<0.01)。FAC-H、FAC-M、FAC-L组小鼠TI、SI、K、A与MG组比较,差异有统计学意义(P<0.05);K、A与VE组比较,差异亦有统计学意义(P<0.05)。结论FAC具有抗衰老作用,能显著提高衰老小鼠血中SOD活性,降低MDA和NO水平,增加衰老模型鼠的免疫器官湿重指数,提高巨噬细胞吞噬功能及淋巴细胞转化率。其作用可能与增强非特异性免疫功能有关。Objective To study the effects of flavonoids of astragali complanalli (FAC) in sub - acute aging mice caused by D - galactose. Methods Sixty ICR mice were randomly divided into groups normal control ( NG group) , model con- trol (MGgroup), VitaminE (VEgroup, 25 mg- kg-~ ~ d-l), FAChigh-dose (FAC-Hgroup, 200mg. kg-~ ~ d-l), FACmedium-dose (FAC-Mgroup, 100mg'kg-~ "d-~) and FAC low - dose (FAC-Lgroup, 50mg·kg-1·d-1), 10 in each. Sub - acute aging mice models were established by injection of D - gal ( 120 mg·kg-1·d-1, 30 d) in all groups ex- cept NG group. From days 21 of establishment, groups FAC - H, FAC - M, FAC - L were fed with corresponding concentra- tions of FAC, 10 d. Blood was taken from the eyes 30 min after the last administration, and superoxide dismutase (SOD) activi- ty, malondialdehyde (MDA), nitric oside (NO), and such indicators of immunity as spleen index (SI), thymus index (TI), carbon clearance index (K) and lymphocyte transformation rate ( expressed by A of OD value measured by 570nm). This experi- ment repeated 3 times, the data averaged. Results When compared with NG group, serum SOD activity decreased, MDA, NO increased in MG group, the difference was significant (P 〈0. 01 ). SOD activity was significantly higher, MDA, NO lower in groups FAC - H, FAC - M, FAC - L than in group MG ( P 〈0. 01 ). There was significant difference in TI, SI, K, A be- tween groups MG and NG (P 〈 0. O1 ), and between groups FAC - H, FAC - M, FAC - L and group MG (P 〈 0.05), and in K, A between groups FAC - H, FAC - M, FAC - L and group VE ( P 〈 0.05 ). Conclusion FAC, having anti - aging effects, can significantly increase serum SOD activity, lower MDA and NO, improve immune organ wet weight index and macro- phage phagocytosis and lymphocyte transformation rate. It may be related to the enhancement of non - specific immune function.
