摘  要:溃疡性结肠炎(UC)的病因和发病机制尚不明确.针对MDR1基因敲除小鼠易发UC;MDR1发生C3435T基因变异在UC患者的发生率显著高于正常人;C3435T基因变异可导致P糖蛋白(P-gp)表达下降;而在激素耐药的UC患者其外周血淋巴细胞及肠黏膜上皮细胞P-gp的表达量较药物敏感者及正常对照组高等现象,提出MDR1基因多态性可能和UC的发生、发展以及激素的治疗反应有关的假说,但也有不同的结论.The etiology and pathogenesis of ulcerative colitis remain unclear. It has been reported that multiple drug resistant 1a (MDR 1a) knockout mice are susceptible to developing a spontaneous ulcerative colitis (UC). The incidence of C3435T gene mutation in the MDR1 gene in UC patients is significantly higher than that in healthy controls. The C3435T gene mutation can lead to decreased expression of P-glycoprotein (P-g-p). The expression of P-gp in the peripheral blood lymphocytes and intestinal mucosal epithelial cells is higher in hormone-resistant UC patients than in hormonesensitive UC patients and healthy controls. Thus, a hypothesis was proposed that the MDR1 gene polymorphism may correlate with the occurrence and progression of UC and the sensitivity to hormone therapy in UC patients. However, this hypothesis is still controversial.
