摘  要:目的:本研究拟采用薄层色谱扫描法建立快速测定患者血清中对乙酰氨基酚浓度的方法,为指导临床合理用药和中毒急救提供依据。方法:实验取100μl血清,用蒸馏水稀释10倍,再取100μl稀释血清用乙醚提取,40℃水浴挥干,残渣用乙醇溶解点样,展开后以λ测245nm,λ参310nm薄层扫描测定。结果:本法板间测定误差小,回收率高,线性范围宽,能准确测定患者血清浓度。结论:这一方法与文献所记载的HPLC和GC相比,具有简便快速、费用低和不需要做衍生化的特点。Objective:The purpose of the study is to establish a method for monitoring serum concentration of acetaminophen with thin layer chromatography scanning method(TLCS),and therefore to guide reasonable administration and treatment for cases who are poisoned with acetaminophen;Methods:This experiment used 100 μl serum of patients with acetaminophen poisoning and diluted to 1 ml,finally 100 μl diluted serum were taken to determinated the serum level of acetaminophen.The procedure was that the first step was the extraction with ether and evaporation in 40℃ water bath, the second was to scan the spots at the wave length of λ245 nm and 310 nm on the thin layer plate after spoting and developing.Results:The determination variations among plates were small with higher recoveries, wider linear range TLCS, compared with HPLC and GC, was simple, rapid, and did not need to derivate acetaminophen.It is suitable for emergency analysis and drug monitoring for acctaminophen.
