摘 要:目的观察益肾蠲湿合剂对肾小球系膜细胞(GMC)增殖及细胞外基质的抑制作用。方法将含低、中、高剂量益肾蠲湿合剂的药物血清作用于10%血清-DMEM培养液培养的GMC,分别用噻唑蓝和酶联免疫吸附(ELISA)方法检测细胞增殖和培养液中基质的浓度。结果益肾蠲湿合剂对GMC增殖有明显的抑制作用,并随着剂量的增加作用也逐渐增强(72h低、中、高剂量的抑制率分别为28.3%、40.5%、47.5%)。同时益肾蠲湿合剂对GMC分泌细胞外基质有也明显的抑制作用(高剂量组对FN、LN及ColⅣ的抑制率分别为26.1%、25.4%、22.3%,中剂量组分别为17.4%、20.5%、15.9%;低剂量组分别为10.4%、10.7%、9.3%)。结论益肾蠲湿合剂对GMC的增殖及基质的分泌有明显的抑制作用,这可能是其有效抑制各种慢性肾炎发展的重要原因。Objective To observe the effect of "Yishenjuanshi" traditional Chinese herbs on the proliferation of glomerular mesangial cells (GMCs) and the secretion of extracellular matrix. Methods All herbs in "Yishenjuanshi" were boiled in water and concentrated into different dozes. GMCs were cultured in serum free medium for 48hs and then 10% FSC-DMEM with or without serum containing " Yishenjuanshi" to detect GMCs proliferation and inhibated secretion of extracellular matrix by MTr and Elisa respectively. Results Low, medium and high dose of "Yishenjuanshi" treatment significantly inhibited GMC proliferation when GMCs were cultured for 24, 48and 72 hs and inhibated secretion of extracellular matrix at 72 hs. The inhibition ratios of cell proliferation increased as the doses increased. Conclusion "Yishenjuanshi" treatment inhibits GMC proliferation and secretion of extraeellular matrix.
