逆灸关元穴对自然更年期大鼠子宫HSP70、HSP70 mRNA、SOD、NOS的影响

摘　　要：目的:探讨逆灸关元穴对自然更年期大鼠子宫热休克蛋白70(HSP70)及其信使RNA(HSP70mRNA),超氧化物歧化酶(SOD)、一氧化氮合酶(NOS)的影响及其机制。方法:将112只通过阴道涂片筛查的10月龄SD雌性大鼠随机分为7组,另设3.5月龄1组为对照,每组16只。观察10月龄逆灸关元穴对随后12、14、16月龄子宫HSP70、HSP70mRNA和子宫抗氧化指标的影响。结果:各自然月数组大鼠子宫HSP70、HSP70mRNA表达随着月龄的增长均呈先升后降的变化,子宫SOD、NOS的活性明显降低(P<0.01)。与同自然月龄组相比,逆灸14、16月龄组子宫HSP70、HSP70mRNA表达明显增加(P<0.05或P<0.01)。子宫SOD、NOS的活性增强。结论:逆灸关元穴可以增加外周子宫组织细胞HSP70及其基因的表达,促进组织SOD、NOS活性增强,这可能是该疗法对子宫产生保护作用的机制之一。Objective： To investigate the influence and mechanism of moxibustion on the acupoint of Guanyuan （CV4） on HSP70,HSP70mRNA,SOD,NOS in uterus of natural climacterium rats. Methods： 112 ten month-aged SD female rats examined by vaginal smear were chosen and divided into seven groups. And another 16 three and half month-aged SD female rats were absorbed. Immunohistochemical, immunoturbidimetric assay and hybridiztion in situ and the like methods were applied to observe the effect on HSP70, HSP70 mRNA, SOD, NOS in uterus of 12, 14 and 16 month-aged menopausal rats with preventive accupuncture on Guanyuan point（twice a week,eight weeks continued） in 10th month. Results： In normal group, the expression of HSP70 and HSP70 mRNA in uterus increased at the beginning and descended in the end with the month growing, the activity of SOD, NOS decreased significantly （ P〈0.01 ） . Compared with control group at the same month-old condition, the expression of HSP70 and HSP70 mRNA of preventiver accupunctre group increased obviously （ P〈0.05 or P〈0.01 ） , and the activity of SOD and NOS increased. Conclusion： Preventive accupuncture on Guanyuan point can improve the expression of HSP70 and HSP70 mRNA and the activity of SOD and NOS in uterus. It might be one of mechanisms of protecting uterus with preventive moxibustion.
