健脾疏肝活血方对H22肝癌小鼠ICAM-1基因表达的影响

摘　　要：目的:探讨健脾疏肝活血方对H22肝癌小鼠细胞间黏附分子-1(ICAM-1)基因表达的影响。方法:采用细胞接种的方法建立小鼠H22肝癌动物模型,将小鼠分为正常组,模型组,环磷酰胺组,健脾疏肝活血方低、中、高剂量6组,采用RT-PCR方法检测健脾疏肝活血方对肝癌组织ICAM-1基因表达的影响。结果:健脾疏肝活血方高、中剂量组的血清sICAM-1皆降低;与模型组比较,具有统计学意义(P<0.05,P<0.01);健脾疏肝活血方中剂量组下调ICAM-1的基因表达;与模型组比较,具有统计学意义(P<0.01)。结论:健脾疏肝活血方下调ICAM-1的基因表达是治疗肝癌的分子机制之一。Objective： To discuss the effect of Jianpi Shugan Huoxue Prescription on ICAM-1 gene expression in H22 hepatic cancer mice. Methods： Animal model of H22 hepatic cancer mice were established by inoculating tumor cell. Mice were randomly divided into six groups： the normal group, the model group, Cyclophosphamide group, low, middle and high dosage groups of Jianpi Shugan Huoxue Prescription. The gene expression of ICAM-1 were detected by reverse-transcription polymerase chain reaction （RT- PCR）. Results： Compared with model group, the contents of ICAM-1 in serum in the middle and high dosage groups of Jianpi Shugan Huoxue Prescription both decreased（P 〈 0.05, P 〈 0.01）. Conclusion： Reducing the gene expression of ICAM-1 was one of molecular mechanism of Jianpi Shugan Huoxue Prescription in treating hepatic cancer.
