摘 要:目的采用新型反相手性色谱柱CHIRALCEL OD-RH(纤维素3,5-二甲基苯基氨基甲酸酯涂敷在5μm硅胶上),建立吲达帕胺对映体的反相高效液相色谱拆分方法,为研究吲达帕胺手性对映体药代动力学提供基础。方法在CHIRALCEL OD-RH柱上,考察了流动相组成、流速和柱温对吲达帕胺手性对映体拆分的影响。结果以水-乙腈(1:1,V/V)为流动相,流速0.5ml.min-1,柱温20℃,在CHIRALCEL OD-RH手性柱上成功拆分吲达帕胺对映体。结论吲达帕胺对映体在CHIRALCEL OD-RH手性柱上获得基线拆分。Objective To develop a RP-HPLC method for the enantiomeric separation of indapamide on a new chiral column CHIRALCEL OD-RH (cellulose tris (3,5-dimethylphenylcarbamate) coated on a 5 μm silica-gel substrate) for pharmaceutical study of indapamide. Methods To obtain optimistic method for the enantiomeric separation of indapamide, the inferences of the composition of mobile phase, the flow rate and the temperature on the enantiomer retentivity and selectivity were examined and discussed. Results The optimized chromatographic condition was as follows: CHIRALCEL OD-RH chiral column(150mm×4.6mm,5?m), water-acetonitrile(1:1, V/V) as the mobile phase, at a flow rate of 0.5ml·min^- 1 and detection at the wavelength of 243nm. The column temperature was fixed an 20℃. Conclusions A baseline separation of indapamide enantiomers was achieved Oil a CHIRALCEL OD-RH column.
