摘  要:目的:建立HPLC测定川芎药材和饮片中游离阿魏酸和总阿魏酸的方法,测定不同来源的川芎样品,以期研究其质量评价指标。方法:用甲醇-甲酸(95∶5)和甲醇-0.24 mol.L-1碳酸氢钠水溶液(95∶5)分别提取川芎样品中的游离阿魏酸和总阿魏酸。HPLC采用Alltima C18(4.6 mm×250 mm,5μm)色谱柱,保护柱为C18(4.6 mm×7.5 mm,5μm);以乙腈和1%冰醋酸水溶液为流动相,梯度洗脱;流速为1.0 mL.min-1;柱温为30℃;测定波长为320 nm;进样量为10μL;以外标法测定样品中游离阿魏酸和总阿魏酸的含量。结果:游离阿魏酸和总阿魏酸分别在0.01~0.20μg和0.05~0.46μg呈线性关系,回收率分别为95.7和98.5%(n=6)。32份川芎药材和饮片中总阿魏酸与游离阿魏酸的含量之比为1.03~4.98,平均值为2.38(n=32);1份川芎栽培苓子中二者之比值达19.6。15份从川芎主产区收集的根茎药材中总阿魏酸的平均质量分数为1.42 mg.g-1,按平均值的80%和药材含水量10%计算,则川芎药材中总阿魏酸的含量以干燥品计为1.25 mg.g-1。结论:本方法可测定稳定、具有可比性的阿魏酸含量,操作简便,适合于川芎药材和饮片中游离阿魏酸和总阿魏酸的含量测定。阿魏酸在川芎样品中以游离和结合型共同存在,但以后者为主,并以总阿魏酸的量发挥药效,故应以总阿魏酸为川芎药材和饮片质量评价的指标。Ferulic acid (FA) is one of the main bioactive compounds in Chuanxiong (CX), the dried rhizome of Ligusticum chuanxiong, but its amount in this herb is difficult to determine accurafely. An accurate quantitative determinationmethod was developed to investigate on the available amount of FA i. e. free FA and total FA. Herbal samples were extracted in methanol-formic acid (95∶5) and methanol-0.24 mol·L^-1 sodium hydrogen carbonate in water (95∶5), respectively and then quantitatively analyzed by HPLC method. Thirty three CX samples were quantified on free and total FA. Total FA was found more abundant than free FA with an average ratio of 2. 38 (n=32) in the range of 1. 03-4. 98 in 32 CX herbs, and a highest ratio of 19. 6 was estimated in a rhizome seedling. Results showed that total FA content would be a better marker for the quality assessment of CX herbs. Fifteen CX typical samples were collected from the trueborn cultivating areas in Sichuan province of China. The amount of total FA in these herbs was estimated to be 1. 42 mg·g^-1 (n=15). The proposed limit of total FA in CX samples should not less than 1. 25 mg·g^-1 calculated on the dried basis. It was also found that the level of total FA was related to the quality, processing method and store duration of CX samples.
