虎门合剂对吗啡依赖模型大鼠学习记忆能力及海马内NOS阳性神经元的影响

摘  要:目的:观察虎门合剂对吗啡依赖大鼠行为学及脑内海马区域NOS的影响。方法:采用逐日递增给药方法建立吗啡依赖大鼠模型;行Y-电迷宫实验检测学习记忆能力;用免疫组化法观察海马部位NOS的变化。结果:吗啡模型组大鼠达到学会标准所需训练次数较正常组明显增多(P<0.01),同时海马CA1区、齿状回内NOS免疫阳性神经元的数目亦明显减少。虎门合剂组大鼠达到学会标准所需训练次数较吗啡模型组明显减少(P<0.01),同时海马CA1区、齿状回内NOS免疫阳性神经元的数目亦明显增多(P<0.01)。结论:虎门合剂对吗啡引起的记忆能力损伤具有拮抗作用,可提高吗啡依赖模型大鼠的学习记忆功能。Objective:To observe the effect of Humen-heji on the behavior and NOS in hippocampus of morphine dependent rats.Methods:Rats were administered(ip)with morphine in gradually increasing doses to establish morphine dependent model.The behavior of rats was measured by Y-electric maze and the changes of NOS positive neurons within hippocampus were observed by immunohistochemistry.Results:In Humen-heji group,learning and memory were improved(P0.01),and NOS immune positive neurons increased significantly in CA1 and DG(dentate gyrus)of hippocampus(P0.01).Conclusion:Humen-heji can antagonize against the general memory damage induced by morphine and improve learning and memory.
